中文摘要 | 以1%明膠溶液與esperase在60℃下,於0.1 M磷酸緩衝液(pH 8.0)中(E/S=1/20)水解,測其水解率與抑制ACE(血管收縮素轉化□)活性能力。結果顯示,明膠經過2 hr水解後水解率達8%左右,IC50值為11.6 mg/mL;抑制ACE活性能力則由3.4%大幅上昇至26.7%。水解物(G2)以Sephacryl S-200膠體柱層析,得到波峰G2-1和G2-2,分子量約在12,900及87,00,IC50值分別為8.1 mg/mL和1.1 mg/mL。經胺基酸組成分析顯示,G2-2的疏水性、鹼性及芳香族胺基酸的含量分別為4.6%,6.5%及1.0%,此含量與G2及G2-1相當。 |
英文摘要 | 1% gelatin solution was hydrolyzed with esperase (E/S=l/20) at 60 in 0.1 M phosphate buffer (pH 8.0) for various periods of time, and the degree of hydrolysis and ACE (angiotensin-converting enzyme) inhibition actirity of all the hydrolysates were determined. Results showed that the degree of hydrolysis of the hydrolysates prepared for 2 hr was 8.0%, and that the IC50 value was 1 1.6 mg/mL. Two peaks (G2-1, G2-2) in the elution pattern of G2 on Sephacryl S-200 were observed. The molecular weights were 12,900 and 87,000, and the IC50 values were 8.1 mg/mL and 1.1 mg/mL for G2-1 and G2-2, respectively. The percentages of hydrophobic, basic and aromatic amino acid in G2-2 were 4.6, 6.5 and 1.0 %, respectively, similar to those in G2 and G2-1. |
