中文摘要 | 1%乳清蛋白溶液與esperase在60℃下,於0.1M磷酸緩衝液(pH8.0)中(E/S = 1/20)水解,測其水解率與抑制ACE(血管收縮素轉化?)活性能力。結果顯示,乳清蛋白經過18小時水解後水解率達12.6%左右。14小時水解物之抑制ACE活性達到最高,此時水解液的水解率約為11.7%,IC50值為1.0mg/ml。 14小時的水解物(A14)以管柱層析分離得到2個波峰A14-1,A14-2其分子量範圍分別為43,700及2,300左右,IC50值分別為0.8及3.2mg/ml。 經胺基酸組成分析顯示,A14-1的疏水性,鹼性及芳香族胺基酸的含量分別為22.2%,14.8%及8.9%。此含量與A14相當,但與A14-2相比則明顯較高。 |
英文摘要 | 1% whey protein was hydrolyzed by esperase (E/S=1/20) in 0.1M phosphate buffer (pH 8.0) at 60℃ for various periods of time. The degree of hydrolysis and ACE inhibitory percentage of all the hydrolysates were calculated. The degree of hydrolysis of hydrolysate was 12.6% after 18 hr hydrolysis. The ACE inhibitory percentage reached maximum after 14 hr hydrolysis (A14), while the legree of hydrolysis and IC50 value was 11.7% and 1.0mg/ml, respectively. Two peaks (A14-1, A14-2) of A14 hydorlysate were observed on Sephacryl S-200. The molecular weight were 43,700 and 2,300, while IC50 values were 0.8 and 3.0mg/ml for A14-1 and A14-2, respectively. The percentage of hydrophobic, basic and aromatic amino acids of A14-1 were 22.2, 14.8 and 8.9%, respectively, which were similar to those of A4, but higher than those of A14-2. |
