中文摘要 | 本研究自3個感染了假性狂犬病(pseudorabies,PR)的豬場共採集了230頭母豬的 初乳與血清,以E醣蛋白(glycoprotein E,gE)酵素免疫吸附試驗(enzyme linked immunosorbent assay, ELISA)及病毒中和試驗(virus neutralization test)同時針對初乳 及血清兩種檢體進行PR抗體之檢測,並比較不同檢體之試驗結果。由gE ELISA結果來分析, 初乳及血清兩種檢體獲得完全一致的結果者佔92.1%,不一致者佔7.9%,其中血清陰性初乳陽 性者佔7.0%,血清陽性初乳陰性者佔0.9%。計算兩種檢體gE ELISA結果的Kappa值為0.84, 證明初乳及血清兩種檢體gE ELISA結果之一致性頗高,並且以初乳做為檢體較為敏感。為了 探究其原因,進—步分析gE抗體陽性母豬血清與初乳中之病毒中和抗體力價,發現初乳檢體 之幾何平均中和抗體力價是1:299.7,約為血清檢體(1:59.3)之5倍(p=.0001)。另外,以初乳 做為檢體進行gE ELISA之重複性可達99%,樣本也易於儲存與運送,本試驗的結論證實可以利 用初乳取代血清做為母豬的輔助檢體用於檢測PR gE抗體。 |
英文摘要 | The aim of this study was to investigate whether colostrum couid be used as a supplementary sample in detecting antibodies against glycoprotein E (gE) of pseudorabies virus (PRV). Serum and colostrum samples were simultaneously taken from 230 sows from 3 farms of pigs endemically infected and vaccinated against PRV. Consistent gE ELISA results, both positive or both negative, found in serum and colostrum samples accounted for 92.1% of the total samples. In contrast, the other 7.9% of the total samples showed discrepant results. Samples negative in serum but positive in colostrum accounted for 7.0%, and samples positive in serum but negative in colostrum accounted for 0.9%. The Kappa value was 0.84, which could be interpreted as a very high consistency between serum and colostrum samples. The mean virus neutralizing titers in colostrum (1:299.7) were about 5 times higher (p=0.001) than for serum (1:59.3). The repeatability of the gE ELISA applied to colostrum was very good, at 99%. Moreover, colostrum is easier to collect, store, and transport under field conditions. It is concluded that colostrum can be used as a supplementary sample in serological screening against PRV in sows. |
