中文摘要 | 從臨床白血病牛隻之腫瘤團塊細胞接種MDBK細胞後分離一株(K1株)牛第四型□疹 病毒(bovine herpesvirus type 4, BHV-4/K1)。將BHV-4/K1以核酸限制□EcoRI與Hind Ⅲ 分別切割DNA分析,結果與國外流行株相似,但其終端重複DNA(polyrepetitive DNA)之EcoR I 片段大小為2.1 Kb,與國外流行株大小不同,可供為毒株區別用,終端重複DNA之單位數目在 株內之每一病毒分子間會有多寡不同之差異,重複DNA之單位最多可達20-25個。從感染 BHV-4/K1之MDBK細胞抽取凝膠沉澱法抗原其方法操作簡單,以此調查本省北部與中部4縣 240頭牛血清,陽性反應者56頭,陽性率23.3%。 |
英文摘要 | A strain of bovine herpesvirus type 4 (BHV-4), K1, was isolated from a cattle with leucosis by innoculating tumor mass into MDBK cells. The DNA fingerprint is similar to foreign strains as compared by restriction endonuclease mapping with restriction endonuclease EcoR I and Hind Ⅲ. The EcoR I fragments of polyrepetitive DNA segment at BHV-4/K1 genome termini, however, is different from the foreign strain by a size of 2.1 Kb in K1 strain. The differentiation of BHV-4 strains can therefore be achieved by DNA fingerprinting. The number of terminal repeat units varies between individual molecules of BHV-4/K1 strains, and can reach as high as 20 to 25 units per DNA molecule. Agar gel precipitation test was used to investigate antibodies against BHV-4 in cattle from north and cential Taiwan. Fifty-six out of 240 cattle (23.3%) were seropositive. |
