中文摘要 | 從濕地松成熟胚切離之子葉培殖體,置於含BAP或zeatin 的BLG培養基中,可誘 導不定芽發生。本研究檢測了細胞分裂素濃度及處理時間對不定芽體誘導及伸長之影響。以 44.4uM BAP 處理培殖體可誘導出最多的芽體,但是短小而未見伸長的芽體,其比例隨著 BAP 濃度增加而增加。 添加 114 及 228uM zeatin 的處理,效果均不及 44.4uM 的 BAP。 芽體誘導及伸長最佳的處理時間均為 14 天。在誘導處理之後,以 BLG 培養基添加 0.5%(v/v) 活性炭繼代培養,可顯著的促進其伸長。在芽體長度及於 4.0 cm 時,以不同濃 度之 IBA(5--270 uM 刺激後再移往不含 IBA 的 1/2GD �粥鷎i基中培養, 可使芽體長出不 定根。 使用濃度為 125 uM 之 IBA 可得最佳發根比例 (89%)。發根之芽體在溫室噴霧系統 下健化並完全成活。 |
英文摘要 | Initiation of adventitious shoots from excised embryonic cotyledons of slash pine was achieved on BLG medium supplemented with BAP or zeatin. The influence of cytokinin concentration and exposure time on shoot initiation and elongation was investigated. Maximum shoot production was obtained at 44.4 uM BAP while the proportion of small nonelongated shoots increased with increasing BAP levels. Zeatin, tested at 114 and 228 uM, was not as effect as BAP at 44.4 uM. A fourteen day pulse treatment with 44.4 uM BAP was best for shoot initiation and elongation. Subsequent to initiation, the use of BLG medium with 0.5% (v/v) activated charcoal enhanced shoot development and elongation. Shoots further elongated to size of 4.0 cm were rooted when pulse incubated with 5 to 270 uM IBA and subsequently transferred to PGR-free 1/2 GD �� medium. An IBA pulse at a 125 uM concentration achieved a best rooting of 89%. All rooted plantlets survived after transfer to a greenhouse with mist system. |
