中文摘要 | 以通常方法進行Nicotiana repanda × N. tabacum種間雜交,雜交後約十二天 花苞變黃脫落,無法得到種子。將授粉後 2、3、5 及 7 天雜交胚珠,利用光學顯微鏡觀察 石蠟包埋染色法處理後之胚珠組織切片。 發現授粉後 2 天之胚珠,已進行雙重受精。第 3 天之胚珠,已可見初期球狀胚,第 5 天者可見完整之球狀胚。但第 7 天時胚與胚珠已開始 出現收縮退化、同時發現所有雜交胚珠,其胚乳均未發育,顯示雜交種子不稔與胚乳發育不 正常有關。 |
英文摘要 | In the conventional crossing between Nicotiana repanda and N. tabacum, it is difficult to obtain interspecific viable seed. The seeds were empty. In order to study the factors causing this empty seed, whole placentae with intact ovules of hybrid were conducting a cross section examination under light microscope on day 2、 3、5 and 7 days after pollination. From the present study, ovule seeds have zygote and visible two polar-nucleus two days after pollination, that is thought to be fertilized. On day 3 and day 5 after pollination, pre-embryo at 8-cell or 12-cell of early globular embryo were observed in the seed. On day 7, few embryos have reached to globular stage, and continuing their development. The collapse of integument was observed in the developed ovules, where corpse of embryo remained. It is considered that the fertilized ovules have to be separately transferred onto a fresh medium to rescue the hybrid embryos. During the course of embryo development, development of endosperm was never found in this study. |
