英文摘要 | Bacillus subtilis isolated from soil produced starch-degrading amylase on triticale, corn, rye, wheat, and soluble potato-derived starches as the carbon source with prior gelatinization. Extracellular amylase produced by Bacillus subtilis showed a single activity band on polyacrylamide gel electrophoresis. The enzyme has a molecular weight of 50 kDa and showed an isoelectric point of 4.9. Optimum pH and temperature for the amylase activity were 6.0-7.0 and 60 ℃, respectively. Extracellular amylase production was increased as cell growth increased. There was no influence on amylase production by medium containing starch and glucose although the amylase production was repressed by medium containing glucose. Furthermore, the amylase production was influenced by the concentration and kind of starches. However, the degradation of starch by amylase was not influenced by the source of starches. |
