中文摘要 | 本研究探討發酵槽中具有剪力的環境下,利用Rhodococcus equi No.23生產膽固 醇氧化脢,培養基組成份之起泡特性,並針對數種除泡劑之適合性進行評估。結果發現所測試 培養液之組成份中,酵母抽出物和 Tween-80 乃為起泡的主要成份。其中以 Tween-80 的起 泡能力最高,而酵母抽出物的泡沫穩定性最佳。在發酵槽中,以液面下通氣的方式進行生產 膽固醇氧化脢時, 雖添加除泡劑並不能完全抑制泡沫的產生, 但若添加 0.4%(v/v)Antifoam-289 時則有利發酵操作之進行, 避免溢流之發生。 在培養液中添加 Antifoam-289,採用液面下通氣培養時,膽固醇氧化脢活性最高點可達約 0.25unti/mL。而 採用液面上通氣培養時,酵素活性最高點只有約 0.11unit/mL。 |
英文摘要 | The foaming property of some componets in medium for the production of cholesterol oxedase using Rhodococcus equi No. 23 and the effectiveness of an antifoam agent in a fermenter were investigated. It was found that yeast extract and Tween-80 were the major components responsible for foaming in the culture medium. Foam initiated by Tween-80 was thicker while the foam formed from yeast extract was more stable. When cholesterol oxidase was produced in a fermenter with submerged aeration, the addition of antifoam agents could not completely repress the formation of foam. However, the addition of 0.4%(v/v) Antifoam-289 could prevent overflow in the fermenter and improve the fermentation process. With the addition of Antifoam-289 and submerged aeration, a maximum cholesterol oxidase activity of 0.25 unit/mL could be achieved in the culture of R. equi no. 23, while no more than 0.11 unit/mL could be obtained in the culture with only head space aeration. |
