英文摘要 | Ketoconazole (KCZ), an imidazole antifungal agent, has a characteristics of causing androgen deficiency. This study was undertaken to investigate the effects of KCZ on steroid hormone secretion of adrenal glands and testes in male dogs. Six healthy male dogs were administered with a high dose (30 mg/kg of body weight/day) of KCZ. Serum concentrations of testosterone, cortisol and progesterone were measured using ELISA kits (NEOGEN E ). Serum biochemical values of AST, ALT, ALP and cholesterol were measured. Numbers of spermatozoa were counted after semen collection every other day. High-dose KCZ significantly decreased serum levels of testosterone, cortisol and cholesterol concentrations (P<0.001). While significantly increased progesterone concentrations (P<0.001). However, the serum biochemical values of AST, ALT and ALP were in the normal range. Oligospermia was observed occasionally in each experimental dog. The results showed that high-dose KCZ inhibited conversion of progesterone to testosterone and cortisol in male dogs. |
