中文摘要 | 本研究之目的,為建立一套非侵入性(non-invasive)、方便、安全且無緊 迫效應之糞便中睪固酮檢測方法,藉以了解禽類生殖內分泌狀況,以改善動物園中野 生鳥類繁殖效率。試驗分三部分:試驗一、建立睪固酮之酵素免疫分析法。試驗二、 以雞為模式,進行血液與糞便性類固醇內泌素濃度相關性研究。試驗三、應用於野生 鳥類,了解繁殖季節與非繁殖季節間,其糞便性類固醇內泌素之變化。結果顯示:本 方法是採用固相競爭型酵素免疫分析法,利用睪固酮-牛血清白蛋白聯結體(T-3- CMO:BSA)免疫兔子獲取抗體;以蕖草根過氧化(horseradish peroxidase)與 T-3-CMO聯結作為標幟抗原,建立睪固酮之酵素免疫分析法。其靈敏度高達2pg/ well;分析內變異C.V.=6.7%,分析間變異C.V.=10.9%;回收率為94.7±13.6 %;且與放射性免疫分析法具極高之相關性(EIA=1.03RIA+28.18,R =0.87)。 雞血液與糞中睪固酮濃度之相關性(R )為0.76。在繁殖季節與非繁殖季節中,雄 藍腹 糞中睪固酮濃度分別為57.6±15.2ng/g與20.0±4.8ng/g,兩者有顯著差 異;而在綠頭鴨也有同樣之變化,其睪固酮濃度分別為79.5±36.0ng/g與27.4± 9.4ng/g。綜合本試驗結果顯示,藉由酵素免疫分析法測定糞中睪固酮濃度,可準確 偵測禽類生殖內泌素之季節性變化情形。 |
英文摘要 | This study was conducted to establish enzyme immunoassays of testosterone in feces. In order to understand the reproductive status and sexing of avian species for improving the reproductive efficacy of captive birds in zoo, this study was designed for the following investigations including: 1. to estab- lish a testosterone enzyme immunoassay, 2. to find the correspondence between fecal and plasma levels of sex steroids in hens, 3. to assess seasonal reproductive function in wild birds. A solid-phase competitive enzyme immunoassay was carried out on microtiter plates. Antisera were obtained from rabbits immunized with T-3-CMO: BSA. A homolgous horseradish peroxidase-linked-testosterone conjugate was prepared by using mixed anhydride procedure. The sesitivity of this assay was 2pg/well. The coefficients of variation of intra and inter-assay were 6.7% and 10.9% respectively. The recovery rate of the system was 94.7 ± 13.6%. The correlation of plasma testosterone concentrations between EIA and RIA was good (EIA=1.03 RIA+28.18, R=0.87). The correlation coefficient between fecal and plasma testosterone was 0.76 (R ) In breeding season (March), the average fecal T level in male Swinhoe's pheasant (Lophura swinhoii) was higher (57.6 ± 15.2ng/g) than those male in the non-breeding season (October)(20.0 ± 4.8ng/g). The Mallard (Anas platyrhynchos) showed the same pattern (breeding season, 79.5 ± 36.0 ng/g; non-breeding season, 27.4 ± 9.4ng/g). In summary, these data indicated that a non-invasive method of fecal testosterone could be applied to assess seasonal reproductive function. |
