中文摘要 | 本研究之目的在嘗試開發出一個簡單和快速檢測飼料中salbutamol方法。先以市 售檢測尿液中心β -agonist 的連接酵素檢驗套組進行樣品篩選工作,對於疑陽性樣品再搭 配以高效液相層析法進行檢測。樣品僅須經簡單地拍提後,通過固相萃取管柱即可進行分析 , 利用 Cosmosil 5C18-MS column: 以 10% 甲醇溶液加 0.1% 三乙基胺緩衝液 (利用磷酸 調 pH 至 3.5) 為移動相,於螢光檢測波長 EX 275 nm 和 EM 305 nm 下進行分析,可成功 地自飼料中檢測出 sa1butamol。平均回收率可高達 94.1%。 |
英文摘要 | The aim of this study is to develop a simple, rapid, and reliable method for determination salbutamol in feeds. Samples were performed using enzyme-linked immunosorbent assay (ELISA) as a screening method. Positive samples were confirmed by high performance liquid chromatography. Salbutamol in the sample was extracted with water (pH 8.8), followed by a Sep-pak C18 (solid phase extraction) clean-up and a high performance liquid chromatography as the means of measurement. The HPLC separation was carried out on a Cosmosil 5C18-MS using methanol-0.1@/otriethylamine (10:90 V/V, pH 3.5 adjusted with o-phosphoric acid ) as the mobile phase. A fluorescence detector was operated at the excitation wavelength of 275 nm and the emission wavelength of 305 nm. This study revealed that salbutamol in feeds could be detected by the modified HPLC method developed by us in 8.7 min. Average recovery of this HPLC method was 94.1%. Therefore, our method is simple, rapid, and reliable for determination of salbutamol in animal feeds. |
