中文摘要 | 本試驗之目的在開發一理想之山羊精液與胚之冷凍保存方法俾提供台灣黑山羊生 殖細胞長久保存之使用,達到保種之目標。 公羊之精液經假陰道方法採出後,比較稀釋前精漿去除與否,及應用蛋黃枸椽酸鈉與脫 脂奶粉兩種不同稀釋液,對山羊精子經冷凍-解凍後之存活率及活力之影響。結果顯示,山 羊精液在稀釋前先行去除精漿,並採用脫脂奶粉為稀釋液者,以及以含有7%甘油為冷凍保 護劑等條件,可獲得理想之冷凍效果,解凍後精子之存活率平均為64.3%,活力為3.8左右, 顯著較其他處理所得之結果為佳(P<0.01)。冷凍保存之精液經解凍後供進行母山羊人工授精 之用,獲得之受胎率為52.6%,受精率則為61.9%,證實所保存之冷凍精液確實具有受精能 力。 由超級排卵母山羊子宮內沖洗出之6~7日齡羊胚,經以含1.8M乙二醇(ethylene glycol) 或10%甘油(glycerol)為冷凍保護劑之保護液,進行慢速降溫冷凍保存,於解凍之後移置於經 發情同期化處理之雜種受胚母羊,分別獲得46.2%及30.0%之受胎率。迄今,種原保存計畫 總計冷凍保存9頭公羊之精液2,035劑,以及山羊胚182個。 |
英文摘要 | This experiment was conducted to examine the methods for goat semen and embryo freezing, and for long term preservation of semen and embryos of Taiwan Black Goat. Fresh semen were collected by artificial vagina. Effects of diluent systems, skim milk and egg yolk-citrate, and removal of seminal plasma before dilution on post-thawed semen quality were examined. Removal of seminal plasma before addition of skim milk diluent and the final diluents containing 7% of glycerol resulted in the optimal post-thawed semen quality (P<0.01), i.e. 64,3% of sperm viability and 3.8 of motility score. The conception rate of estrus synchronized does and the fertilization rate of ova in superovulated donors inseminated with the frozen-thawed semen were 52.6% and 61.9%, respectively. Embryos recovered from donors of Taiwan Black Goat were frozed along with the frozen-thawed semen were 52.6% and 61.9%, respectively. Embryos recovered from donors of Taiwan Black Goat were frozen along with the solution containing either 1.8 M of ethylene glycol or 10% of glycerol by conventional slow freezing method. Ethylene glycol was superior to glycerol as a cryoprotectant for goat embryo freezing while evaluated by the conception rate of recipients in an embryo transfer trial (46.2% vs 30.0%). A total of 2,035 straws collected from 9 bucks and 182 embryos have been preserved so far. The results in this experiment demonstrated that these frozen semen and embryos could be preserved as an applicable gene pool of Taiwan Black Goat for further application. |
