中文摘要 | 豬胚冷凍保存將有助於豬種基因庫之建立和豬胚移置工作國際化之拓展。本試驗 把豬胚置於含 1.5 M 甘油冷凍保護劑之麥管中, 自 25 ℃起以 1 ℃ / 分的速率降溫到 -6.8 ℃,維持 10 分鐘的植冰後,改以 0.3 ℃ / 分的速率降溫到 -35 ℃,停留五分鐘後 再將麥管直接置入液態氮中保存。 解凍時將麥管自液態氮中取出並迅投入 37 ℃水浴溫 20 秒,使之融解並以 0.3M 蔗糖溶液來透析出甘油後,進行鏡檢和 24 小時體外培養。冷凍胚 存活率是以培養後可進一步發育的胚數所佔冷凍胚數之百分比表示之。試驗一比較自單細胞 、雙細胞、四細胞、八細胞、桑椹、早期囊胚、擴張囊胚到孵化囊胚期的蘭嶼豬胚冷凍存活 率,分別為 19.2 (5/26)、0.0(0/58)、6.8(3/44)、6.5(3/46)、26.3(5/19)、5.9(1/17)、 36.1(13/36) 和 75.6%(34/45)。 試驗二比較蘭嶼、藍瑞斯和杜洛克種的擴張囊胚之冷凍存 活率,分別為 37.3(19/51)、5.4(6/110) 和 9.4%(5/53); 以及比較蘭嶼、藍瑞斯和約克夏 種的孵化囊胚之冷凍存活率,分別為 51.5(34/52)、21.4(3/14) 和 0.0%(0/10)。存活並發 育為擴張囊胚者經螢光染色後,其粒線體分佈呈現均勻或僅局部不均勻,大多數胚葉細胞核 呈現頗一致的核相,本試驗結果顯示冷凍胚存活率有胚期與品種之差異存在。 |
英文摘要 | Cryopreservation of porcine embryos will help the establishment of embryo banking for pig genetic resources and the promotion of international movement of embryo transfer. Embryos were incubated at 1.5 M glycerol culture medium prior to cooling. The cooling rate was set at 1 ℃ /min from 25 ℃ to -6.8 ℃. At -6.8 ℃, embryos were maintained for 10 minutes for seeding and then the freezing rate was reset at 0.3 ℃ /min from -6.8% to -35 ℃. After five minutes of setting at -35 ℃,the straw containing frozen embryos was directly put into liquid nitrogen for preservation. Thawing of embryos was made by putting the frozen straw into the 37 ℃ water bath directly for 20 seconds. Embryos were diluted in 0.3 M surcose gradient solution to remove glycerol and then cultured for 24 hours to examine their development morphologically. The survival rate of embryos was designated was the percentage of thawed embryos, having a normal development to the subsequent cell stage, to the number of frozen embryos. Experiment I: Embryos from 1-, 2-, 4- and 8-cell, morula, early blastocyst, expanding blastocyst to the hatched blastocyst of Lanyu pigs were frozen to test the embryonic freezability at various cell stages and they developed 24 hours after thawing with survival rates of 19.2 (5/26), 0.0 (0/58), 6.8 (3/44), 6.5 (3/46), 26.3 (5/19), 5.9 (1/17), 36.1 (13/36) and 75.6% (34/45), respectively. Experiment II: Breed comparison on survival rates of expanding blastocysts of Lanyu, Landrace and Duroc pigs was conducted. There were 37.3 (19/51), 5.4 (6/110) and 9.4% (5/53), respectively, of thawed embryos having normal cell development. Survival rates of Lanyu, Landrace, and Yorkshire hatched blastocysts were 51.5 (34/52) , 21.4 (3/14) , and 0.0% (0/10) , respectively. Embryos survived and developed to the expanding blastocyst stage were fluorescently stained. Mitochondrial distribution were homogeneous in some embryos although several embryos were less homogeneous or partially homogeneous. Most of nuclei of blastomeres had a uniform fluorescent stainging. In summary, there were differences in the freezability of porcine embryos among breeds and cell stages. |
