中文摘要 | 胚幹細胞係分離自哺乳動物埋植前階段的胚,乃為一種且具有多能性分化能力的 細胞。株化胚幹細胞之成功要件,端視此等細胞是否存在於發育中的胚體,以及分離出的細 胞是否能在體外培養的條件下繼續分裂,同時仍能維持未分化的狀態。本研究的目的,旨在 探討最適的豬胚發育階段,以利豬胚幹細胞的分離與株化。試驗係將自桑椹胚到晚期孵化囊 胚各發育階段的豬胚,分別與小鼠胎兒成纖維細胞(STO cells)製備的供養層細胞共培養在 DMEM添加核 酸、β-羥基乙硫醇、抗生素、與胎牛血清的培養液中。並於適當時機將衍 生之細胞群落予以分離與繼代培養,依其細胞表型與分化能力之表現予以株化。試驗結果顯 示,孵化前的豬胚與中、晚期孵化囊胚在此共培養系統中,無法分離、株化出穩定而未分化 的胚衍生細胞株。而在相同的共培養系統之條件,從早期孵出囊胚則分離株化出12株胚衍 生細胞株。此等細胞株當中具有三株表型穩定、正常之核型、並表現多能性分化能力等與小 鼠胚幹細胞相似的特性。 |
英文摘要 | Embryonic stem (ES) cells are pluripotent cells isolated from in vitro culture of preimplantation embryos. Presence of pluripotent cell population in developing embryos and the possibility of these cells to be further proliferated in vitro without differentiation determine the success in isolation and establishment of ES cell lines in pigs. The objective of this study was to determine the most appropriate developmental stages of the porcine preimplantation embryos form isolation and establishment of porcine ES cell lines. Whole embryos from morula to late hatched blastocyst stages were co-cultured at 39℃ with meiotically inactivated permanent murine embryonic fibroblasts (STO) in Dulbecco's modified Eagle Medium supplemented with nucleotides, mercaptoethanol, antibiotics, and fetal bovine serum. Of pre-hatched porcine embryos, co-culture with STO feeder layers did not result in any stable, undifferentiated embryonic cell lines. On the other hand, twelve undifferentiated porcine embryo-derived cell lines were established from intack embryos at the early hatched blastocyst stage. Three of them had stable morphology, normal euploid porcine karyotype, and shared some other typical characteristics with murine ES cells. |
