中文摘要 | 細胞凋亡(apoptosis)為一種獨特的細胞死亡形式;其形態上主要變 化包括細胞膜泡狀囊(bleb-bing)的形成,細胞核內染色質聚集(chromatin condensation),去氧核醣核酸斷裂(DNA fragmentation)以及細胞凋亡體 (apoptotic body)的形成等特徵。細胞凋亡發生與調控的途徑,目前雖然並不 完全清楚;然而不同原因所誘發之細胞凋亡,其調控的機制或許不同;但最後其 胞內鈣離子濃度皆會上升,進而活化去氧核醣核酸水解酵素(endonuclease), 造成去氧核醣核酸斷裂。由於胞內鈣離子濃度主要由內質網與粒線體調控;而與 細胞凋亡有密切關係的Bc1-2不但位於內質網與粒線體的膜上,同時也與胞內 鈣離子濃度的調控有關;這些現象說明可經由粒線體和內質網來調控胞內鈣離子 的濃度,進而調控細胞凋亡。最近,本實驗室發現視網酸(retinoic acid)和 phorbol ester皆可誘發人類子宮頸癌HeLa細胞進行細胞凋亡;而在細胞凋亡 前,其內質網與粒線體內的鈣離子皆大量釋出。另外,以thapsigargin釋出內 質網內的鈣離子;或以ionomycin釋出粒線體與內質網內的鈣離子也可誘發HeLa 細胞進行細胞凋亡。以上結果進一步證明可經由粒線體和內質網來調控胞內鈣 離子的濃度,進而調控細胞凋亡。 |
英文摘要 | Apoptosis, one of well-recognized modes of cell death, is usually defined by plasma membrane blebbing, cell volume reduction, chromatin condensation, DNA cleavaged into nucleosomal multiples and formation of apoptotic bodies. Although the cause and execution mechanism are not clearly understood, activation of endonuclease by high concentration of intracellular Ca□ is a common phenomena in apoptosis induced by various factors. This paper reviewed the evidence which indicated intracellular Ca□ played the critical role in apoptosis and was regulated by a variety of mechanism existed in mitochondria and endoplasmic reticulum. In recent studies, we found that retinoic acid and TPA released Ca□ from mitochondria and endoplasmic reticulum, before induced apoptosis in human cervical cancer HeLa cells. In addition, thapsigargin (release Ca□ from endoplasmic reticulum) and ionomycin (release Ca□ from endoplasmic reticulum and mitochondria) both induced apoptosis in HeLa cells. These results further support that abrupt release of Ca□ from endoplasmic reticulum and mitochondria is readily to trigger apoptosis. |
