中文摘要 | 粒線體外膜的VDAC (voltage dependent anion-selective channel) channel被認為是粒線體對外的唯一孔道,因此能夠控制 VDAC 開關的物質勢必能調節粒線體的功能。在粒線體的膜間隙存在的一種蛋白質稱為調節者,對 VDAC 的通透性具有強烈的抑制作用,因此被認為其對粒線體的功能具有調節作用,但此蛋白質尚未被純化出來。本試驗的主要目的,仍在建立一套新穎的方法來檢測此蛋白質的活性,其原理為調節者抑制 VDAC 的通透性,勢必減低完整粒線體內膜酵素的活性,利用內膜酵素活動減弱的程度作為調節者活動的指標;本試驗同時發現離子強度會影響調節者的活性,在10mM KCl環境下,無法測得調節者的活性,但在160 mM KCl的環境下,調節者表現出強烈的活性。 |
英文摘要 | VDAC channels on the mitochondrial outer membrane are recognized as only pathway for mitochondria to external environments. The agents, therefore, which can control VDAC gating mechanisms should regulate mitochondrial functions. The modulator, a soluble protein located in the mitochondrial intermembrane space, could largely reduce the permeability of VDAC. Therefore, it is considered to be a candidate to regulate the mitochondrial function. This protein have yet been purified. The main goal of this study is to establish a novel method for detecting the enzymatic activity of the modulator. Basically, the modulator should decrease activities of enzymes on the inner membrane of intact mitochondria if it inhibits the permeability of VDAC on the mitochondrial outer membrane. The degree of a decrease in the activity of the mitochondrial inner membrane enzyme could be used as an indicator for the activity of a modulator. This study also finds that ionic strength can affect the activity of the modulator. Nothing could be detected for the activity of the modulator in the presence of 10mM of KCl. However, the activity of the modulator is very stron in the presence of 160mM of KCl. |
