中文摘要 | 研究已證實當生物體受到外界刺激而產生痛覺後,某些神經原內c-fos proto- oncogene 的活性會上升, 因此利用免疫細胞化學法來辨識 c-fos proto-oncogene 的表現 ,可作為神經原活性之指標。蟾酥為中國民間應用於強心、止痛、止血等的常用藥物。經成 份分析後,知其中的 bufalin 為其主要藥效成份。 本研究在分析以 formalin 誘導天竺鼠 產生痛覺後, 再注射 bufalin,以觀察動物脊髓背角區神經 c-fos proto- oncogene 之變 化。 天竺鼠於接受 0.lml l% formalin 之注射後 (注射部位為後足 ),可於腰脊髓節觀察 到有 c-fos proto-oncogene 免疫及應之細胞。實驗組之動物於接受 formalin 之注射前或 後 30 分鐘,接受腹腔內注射 0.lml 0.005% bufalin, 其腰脊髓節還是可觀察到有 c-fos 免疫反應之細胞, 其數量較對照為少, 但數量之差異在統計上不顯著。 本研究證實 formalin 可誘導痛覺之產生, 因而引起動物脊髓背角區神經原 c-fos proto-oncogene 之 表現,而實驗之 bufalin 濃度對痛覺之抑制則不明顯。 |
英文摘要 | C-fos is a proto-oncogene that is expressed within some neurons following peripheral stimulation. The protein product, c-fos protein, can be identified by immunocytochemical techniques, therefore, c-fos expression might be used as a maker for neuronal activity. Bufalin, an active principle of the traditional Chinese medicine chan'su, has been proved to be a potent pain-releaser. The present study has analyzed patterned of c-fos expression in the neurons of the dorsal horn in the spinal cord of both control and experimental animals subjected to formalin induction and injection of bufalin before or after formalin injection. Cells with c-fos immunoreactivitY was found in the lumbar spinal cords of the control animals. Labelled cells were also found in the lumbar spinal cords of the experimental animals. The |
