英文摘要 | Effect of acupuncture in Zusanli on pain sensation in relation to the changes of cerebral opioids was investigated in rats using tail-flick method. Antinociceptive action was observed in rats that received an electrostimulation via acupuncture inserted in Zusanli point for 30 min in a way dependent on stimulatory frequence from 1 Hz to 100 Hz. Naloxone at the dose sufficient to block opioid receptors abolished the antinociceptive action induced by electrostimulation at 1 Hz and 10 Hz and attenuated the response to electroacupuncture at 100 Hz. In the meantime, plasma β-endorphin was also markedly elevated in rats receving electroacupuncture in a frequence-dependent manner. However, change of plasma β-endorphin was not significant in adrenalectomized rats that possessed an antinociceptive action by electroacupuncture in Zusanli point for 30 min. It is indicated that increase of plasma β-endorphin by electrostimulation in Zusanli point can be considered as a stress response wich is related to adrenal gland but is unrelated to antinociceptive action. Radioimmunoassay of opoids in brain and spinal cord shows an elevation of enkephalin-like immunoreactivity (Enk-ir) in hypothalamus and striatum from rats that received an electrostimulation in Zusanli point for 30 min. Increase of dynorphin-like immunoreactivity (Dyn-ir) was observed in spinal cord from the rat receiving stimulation at 100 Hz for 30 min. Moreover, β-endorphin-like immunoreactivity (BE-ir) was raised in the brain from rats that received acupuncture for 30 min. Northern blotting analysis of mRNA for Pre-Pro-Enk in hypothalamus and for Pre-Pro-Dyn in spinal cord from rats that received the electroacupunctrue, no difference can be obtained in samples from rats that received an electrostimulation for 30 min with the non-stimnlated centrol. However, mRNA levels for Pre-Pro-Enk and Pre-Pro-Dyn were increased in tissues from rats that received the repeated electrostimulation every 2h for 4 times and once more in the next morning, 30 min before decatitation. Transient electrostimulation of Zusanli point in the rat induced an incerease of opioids in central nervous system without an activation of transcription for peptide synthesis indicating the possibility that progress of opioids from Pre-Pro-form is facilitated by acupuncture at Zusanli point. This finding is helpful in the research of opioids relation to acupuncture. |
