中文摘要 | 本研究探討養殖的草蝦與草魚之肌漿網膜中以鐵催化的脂質過氧化系統的某些 特性。肌漿網膜由草蝦與草魚的肌肉中分離出.並與含有FeCI3,ADP,NADH或NADPH的反應溶 液混合後產生脂質過氧化作用。本系統以TBA(Thiobarbituric acid)反應物質量作為脂質過 氧化系統的活性指標。當反應溫度由6℃提高到26℃時,不管是草蝦或草魚的脂質過氧化皆 隨溫度提高而上升。然而當反應溫度為66℃時,脂質過氧化則被抑制。當反應PH值在6.0到 7.0之間時,較酸性的環境會提高脂質過氧化的速率。草蝦的肌漿網膜系統中,利用NADH作 為還原劑的能力比NADPH強很多(p<0.05);然而在草魚的系統中,則便用NADH或NADPH為還 原劑所得到的脂質過氧化速率並無顯著差異(p>0.05)。本研究之結果可提供適合研究飼料抗 氧化營養成份在次細胞層級上對於動物體影響的實驗工具。 |
英文摘要 | To characterize the iron-catalyzed NADH or NADPH dependent lipid peroxidation system in sarcoplsamic reticulum (SR), the SR was extracted from grass shrimp and grass carp muscles and incubated with FeCl3, ADP, NADH or NADPH. Production of thiobarbituric acid reactive substances (TBARS), indicating the lipid peroxidation activity in this system, of SR from both species increased with the increase of reaction temperatures from 6°to 26℃. The activity substantially decreased at 66℃ to almost the same level as that at 6℃. It was also found that the lipid peroxidation activity increased with the decrease in pH between 6.0 and 7.0. According to the data obtained in this study, the lipid peroxidation system preferred NADH to NADPH in SR of grass shrimp, while it could use both NADH and NADPH equally as reducing agent in that of grass carp. This study can provide important tools for the dietary anti-oxidative nutrient study at subcellular level. |
