中文摘要 | 牛乳中oestrogen抽取法及利用毛細管氣體色層分析(Capillary GC)測定其中oestrone、oestradiol-17β、oestrdiol-17α、oestriol含量之方法已建立,即牛乳脂肪部分先行皂化,oestrogen部分用乙醚抽取並且濃縮,利用heptaflorobutyric anhydride (HFBA)在丙酮及Chlorotrimethylsilane (TMSC)溶劑中將oestrogen接上HFBA,GC分析中利用電子補獲偵測器(ECD)可檢測這些oestrogen濃度在10 pg/ml左右之各項成分,測定結果牛乳中oestrogen及oestriol大致在2~200 pg/ml及20~120 pg/ml範圍內,而oestradiol-17β,11頭乳牛中只有一頭被測出其濃度是102 pg/ml,而11頭乳牛中oestradiol-17α只五頭被測出其濃度在0~107 pg/ml之間。探討泌乳期中oestrogen變化,oestrogen與oestriol濃度隨著泌乳期間增加而成正比例上昇,到8個月之後變動很大而濃度下降,另oestradiol-17α則泌乳初期濃度很低,第7個月之後才頻頻被測出,其濃度在0~107 pg/ml之間。 |
英文摘要 | Procedures are described for extraction and gas chromatography (GC) of the oestrogen in whole milk. This is accomplished by removing lipid through saponification and ether extraction, and oestrogen was concentrated by evaporation. The oestrogen is derivative with heptaflorobutyric anhydride (HFBA). Using an electron capture detector in GC analysis, the detectable level of oestrogen in milk was 10 pg/ml. The oestrone was in the range of 2~200 pg/ml, and oestriol was 20~120 pg/ml in eleven individual cow milk samples. However, oestradiol-17β was detected in one cow only (102 pg/ml). For the other cows, it was undetectable. Oestradiol-17α was deteded in five cows and with the range of 0~107 pg/ml were not detected in the other cows. The change of milk oestrogen concentration through the whole lactating period was also investigated. The oestrone and oestriol were at low level in the preliminary period but increased when the lactation proceed. At late lactation, their concentration was great in variation. The estradiol-17α is also low in the preliminary period even undetectable until the seven the month. |
