中文摘要 | 馬兜鈴酸是馬兜鈴屬(Aristolochia)植物的主要成分,具有利尿、增強免疫功能、增加吞噬細胞能力及抗腫瘤功效,已開發用於臨床,然因有報告指出,馬兜鈴酸具有誘變性為致癌因子及懷疑與比利時中草藥腎病(CHN)有關,故引起社會大眾的疑慮。本試驗以高效液相層析儀,分析含木通的中草藥方劑內馬兜鈴酸的含量,方劑包括:辛夷散、導赤散、消風散、龍膽瀉肝湯、八正散、當歸四逆湯。本法採用C18逆相層析管柱,移動相為乙�[ : 2 % 醋酸溶液(45:55 v/v),流速為0.8 mL/min,檢測波長為395nm,得到良好的分析結果。馬兜鈴酸Ι在濃度0.63-100.00μg/mL,利用上述分析技術進行對照標準品馬兜鈴酸之分析,得線性迴歸方程式(Y=mX + b)及相關係數(r)為Y=2.83e + 004X + 2.11e + 002(r=0.9999),顯示良好線性關係。馬兜鈴酸I同日內及異日間相對標準偏差為0.23-1.52%及1.26-3.32%,顯示再現性可以接受。添加回收率為88.5-103.0%,顯示準確性可以接受。價購市售濃縮中藥方劑46件,利用上述高效液相層析法分析,共16件含馬兜鈴酸 I,含量介於0.05-2.32 mg/g之間,平均含量則因方劑不同而有差異。 |
英文摘要 | A high performance liquid chromatographic (HPLC) method for the determination of aristolochic acid in traditional Chinese medicinal prescriptions containing Caulis Aristolochiae Manshuriensis was established. The prescriptions included Xin Yi San (辛夷散), Dao Chi San (導赤散), Xiao Feng San (消風散), Long Dan Xie Gan Tang (龍膽瀉肝湯), Ba Zheng San (八正散), and Dang Qui Si Ni Tang (當歸四逆湯). The samples were analyzed using the HPLC method on a LiChrospher 100 RP-18e column and detected at 395 nm with acetonitrile and 2% (v/v) acetic acid (45:55, v/v) as the mobile phase at a flow rate of 0.8 ml/min. The regression equations of aristolochic acid I was Y=2.83e+004X+2.11e+002 (r=0.9999). The intraday and interday relative standard deviations of aristolochic acid I were at the levels of 0.23-1.52%, 1.26-3.32%, respectively. The recoveries of aristolochic acid I added in the six kinds of blank prescriptions were between 88.5 to 103.0%. Forty-six samples of commercial concentrated preparations containing mutong were analyzed and the results showed that 17 samples contained aristolochic acid I. The amount of aristolochic acid I in samples was between 0.05 to 2.52 mg/g. |
