中文摘要 | 對人類嗜T淋巴細胞病毒第一型(HTLV-I)異活化物Tax1的抗體反應以三種方法分 析,其一為放射免疫沉澱法(RIPA),另外兩種則為西方墨跡法(WB)。WB之一是直接購得,另 一則是以僅特異地產生Tax1的HTLV-I非產殖細胞株C81-66-45的細胞解離液自製WB。於23 件HTLV-I陽性血清中,11件為RIPA陽性,而兩種WB都為陽性者有10件。雖然RIPA的敏感 度較高,但是會有非特異性的反應。有四件檢體的結果不一致,其一僅RIPA為陽性,而兩種 WB皆為陰性,另一則兩種WB皆為陽性但RIPA為陰性,剩餘的兩件則RIPA皆為陽性但僅一 種的WB為陽性。最強反應的檢體來自一位HAM(HTLV-I相關脊髓病變)的患者,以自製的WB 可得到2 ×105的力價。亦檢測10件HTLV-Il的血清對Tax1的反應,雖然有一半的HTLV-Il 血清為RIPA陽性,但僅有對HTLV-I所有的抗原皆呈現最高反應的那件血清才為自製的WB 測得陽性,且其力價低得僅有200。對此不一致性加以討論,而因不同的方法導致的抗體反應 差異很值得注意。 |
英文摘要 | Antibody reactivity to the transactivator Tax1 of human T-lymphotropic virus type I (HTLV-I) was assayed by three methods, one was radioimmunoprecipitation assay (RIPA), the other two were Western blot (WB). One of the WB was a commercial kit. The other WB was homemade by using the cellular lysate of an HTLV-I non-producing cell line C81-66-45 which specifically provided the Tax1. Out of 23 HTLV-I positive sera, eleven was positive by RIPA while 10 was positive by the two WBs. Though sensitivity by RIPA was higher, nonspecific reactivity had occurred with it. There were four discordant sera, one was positive by RIPA but negative by the two WBs; another was positive by the two WBs while negative by the RIPA; the rest two were positive by the RIPA but positive only by one of the WBs. The highest reactivity was shown by a HAM (HTLV-I associated myelopathy) patient whose titer reached 2 ×105 by the in house WB. Ten HTLV-II sera were also tested for their reactivities to Tax1. Though half of the HTLV-II sera were positive by the RIPA, only the one that showed the highest reactivities to all the HTLV-I antigens was positive by the in-house WB, and the titer was as low as 200. The discordance was discussed. The difference in seroreactivity resulted from different assay methods should be aware of. |
