中文摘要 | 建立以單一血清稀釋倍數測定鴨血清中定禽霍亂(fowl cholera,FC)之間接酵素結合免疫吸附法(ELISA)。最適當之ELISA反應條件為以超音波擊?菌體製備之抗原1:5倍稀釋,測試血清1:400倍稀釋,酵素標示抗體1:1000倍稀釋,抗原抗體反應時日30分鐘,酵素受質感作時間45分鐘。檢定同一血清在ELISA盤內不同位置所得OD值,發現不同行所得OD值有顯著差異,不同列之間則無差異,經以在相對行之位置進行二孔重覆測定之平均值可矯正之。本ELISA試驗重現性良好,並可用於鴨隻接種FC菌苗後之監測。鴨隻於4日齡接種同一活菌菌苗,於14日齡時再分別以疫苗A或疫苗B油質佐劑疫苗補強,所有供試鶻隻在1及2週齡時皆為ELISA抗體陰性,疫苗A接種組在4週齡時抗體平圾331倍,7週齡時平均2,033倍,疫苗B接種組在4週齡時抗體平均3,278倍,7週齡時平均4,573倍,兩組間有顯著差異(p<0.05)。 |
英文摘要 | An enzyme-linked immunosorbent assay (ELISA) was developed for measurement of anti-Pasteurella multocida antibody titer in ducks in a single serum dilution. The optimum test condition for the ELISA was determined by the ratio of the OD value of the positive control serum to negative control serum(P/N ratio). It was found that 1:5 dilution of the sonicated antigen, 1:400 dilution of test serum, 1:1,000 dilution of a commercial goat anti-duck HRPO conjugate gave best results. The antibody-antigen reaction time of 30 minutes, and substrate-enzyme reaction time of 45 minutes was chosen by the same criteria. In-plate variation was demonstrated among different columns(P<0.05) but not among different rows (P>0.05). The variations among different columns could be corrected by test duplicate samples at the opposite columns of the plate. The ELISA was shown to be reproducible and was used to monitor the antibody response in ducks after vaccination. The antibody response of ducks vaccinated with a live FC vaccine at 4-day-old, and revaccinated again with inactivated vaccine A or B at 14 days of age was examined. Ducks revaccinated with vaccine A had a mean ELISA titers of 331 at 4-week-old, and of 2,033 at 7-week-old. Ducks revaccinated with vaccine B had a mean ELISA titers of 3,278 at 4-week-old, and of 4,573 at 7-week-old. the antibody response of the two groups differ significantly(P<0.05). |
