| 中文摘要 | 以雜桉成熟木經多芽體透導繁殖獲得之試管內小植株為材料,取其節間及帶有葉脈之葉柄做為培植體,結果發現添加200 mg/l醯胺麩胺酸(glutamine)、100 mg/l酪蛋白(casein hydrolysate)、100%椰子汁及1 mg/l BA與3 mg/l NAA組合,或0.05∼0.5 mg/l TDZ之B5無機鹽類培養基可使節間及葉柄形成再生之莖體。抗壞血酸(ascorbic acid)具有促進及加速莖體形成之作用。形成莖體之最佳培養基為含有0.1 mg/l TDZ及80 mg/l ascorbic acid之B5無機鹽類培養基。莖體經移至添加0.1 mg/l BA及0.01 mg/l NAA之MS培養基會快速增殖。若繼代培養於未添加植物生長調節劑之1/2 MS培養基,莖體則會抽長。已抽長之莖體可利用先前發展之方法快速培育成小植株。 |
| 英文摘要 | Mature tree-derived plantlets were produced through in vitro multiple shoot induction. Internode segments and petioles with partial leaf veins from the plantlets were used as explants to regenerate shoots. The first shoots were regenerated from internode segments cultured on B5 medium supplemented with 200 mg/l L-glutamine, 100 mg/l casein hydrolysate, 10% coconut milk and plant growth regulators, 1 mg/l benzyladenine (BA) and 3 mg/l naphthaleneacetic acid (NAA) in combination, or 0.1 mg/l thidiazuron (TDZ) singly. Furthermore, shoot organization was obtained from internode segments cultured on B5 medium supplemented with TDZ concentration ranging from 0.05 to 0.5 mg/l. Shoots were also regenerated from petioles with partial leaf veins cultured on B5 medium containing 0.1 mg/l TDZ. Ascorbic acid was useful in the enhancement and acceleration of shoot formation that was optimal on B5 medium supplemented with 0.1 mg/l TDZ and 80 mg/l ascorbic acid. Shoots proliferated rapidly upon transfer to MS medium containing 0.1 mg/l BA and 0.01 mg/l NAA, and elongated on half the strength of MS medium without plant growth regulator. The elongated shoots readily root to produce plantlets according to the previously developed method. |
