中文摘要 | 本試驗探討影響Lactococcus lactis subsp. Lactis CCRC 14016生成乳酸鏈球菌素的一般重要因子。結果顯示在各種測試碳源中,可溶性澱粉是生產乳酸鏈球菌素最佳的碳源。即在4.0%可溶性澱粉培養下,乳酸鏈球菌素的產量達2,000IU/ml,約為葡萄糖產量的3倍。在可溶性澱粉的培養基下,葡萄糖成為乳酸鏈球菌素生產的一個不利因子。另外,在培養基中添加酵母抽出物、Tween 80,不但可促進菌體的生長,亦可刺激乳酸鏈球菌素的分泌,而在50mM MOPS緩衝溶液系統下,可減緩發酵液pH值下降速率,有利於菌體生長並可提高鈣酸鏈球菌素產量約46%。 |
英文摘要 | This experiment was conducted to investigate some important factors which affected the production of nisin by Lactococcus lactis subsp. Lactis CCRC 14016. The results showed that soluble starch was the best carbon source for the growth and nisin production by L. lactis CCRC 14016. When cells were cultivated in a medium containing 4.0% soluble starch, they produced about 2,000 IU/ml of nisin which was three times higher than a medium using glucose as a carbon source. In soluble starch broth, the presence of glucose was unfavorable for nisin production. Yeast extract ad Tween 80 were found to stimulate cell growth and nisin formation. When cells were grown in a medium buffered with 50 mM MOPS, the growth of cells became favorable and nisin titre was also increased by about 46%. |
