中文摘要 | 近年來,發酵工業快速發展,舉凡抗生素、胺基酸、蛋白質、酵素及生物殺蟲劑等生物性產物,均廣泛應用發酵工程技術,以期達到增加產能且降低生產成本的目的。在本研究中,以蘇力菌達姆斯塔丁斯亞種(Bacillus thuringiensis subsp. darmstadiensis)為生產菌株,以法國巴斯德研究所de Barjac博士提供的蘇力菌素為標準品,經由高效液相層析法分析蘇力菌素之產量;在相同的培養條件下,比較網狀內管氣舉式反應器.氣泡塔及攪拌槽培養蘇力菌生產蘇力菌素之效能,其中攪拌槽以兩種攪拌轉速(500,800 rpm)分別測試,結果顯示,攪拌轉速800 rpm攪拌槽的蘇力菌素僅得1.03 mg/mL最低產量,網狀內管氣舉式反應器則達到最高產量2.22 mg/mL,氣泡塔則因質傳的限制,其產量為1.86 mg/mL低於前述之新型反應器。網狀內管氣舉式生化反應器質傳效率和混合效果良好,適合好氣性蘇力菌生長並分泌高量的蘇力菌素。 |
英文摘要 | Fermentation technology has been widely applied to the industrial production of antibiotics, amino acids, proteins, enzyme, and bioinseticides. In this study, a potential bilinsecticide thuringiensin productions from Bacillus thuringiensis subsp. darmstadiensis using a conventional stirred-tank with agitation rates of 500 and 800 rpm, a bubble column, and an airlift reactor with a net draft tube fermentation systems were investigated. The results showed that fermentation in the strirred-thank with higher agitation rate (800 rpm) produced only 1.03 mg/mL thuringiensin due to the effect of high shear in the system. The modified bubble column-airlift reactor with a net draft tube gave the highest thuringiensin production of 2.22 mg/mL. The conventional bubble column which had limitation of oxygen transfer had less thuringiensin production of 1.86 mg/mL. In conclusion the modified bubble column bioreactor is suitable for culturing aerobic Bacillus thuringiensis and the production of thuringiensin. |
