| 中文摘要 | 肺泡巨噬細胞為肺臟清除入侵之病原微生物或吸入之顆粒的重要細胞,故如能促 使此種細胞的數目及吞噬能力增加,將有助於肺臟預防病原體的感染。曾有報告指出,豬經 假性狂犬病疫苗免疫後,肺泡內巨噬渦胞的數目及吞噬能力,均明顯的增加,本實驗即擬更 進一步的比較油劑或水劑假性狂犬病疫苗,不含佐劑之假性狂犬病病毒顆粒,豬瘟疫苗,放 線桿菌菌苗以及佛氏完全或不完全佐劑免疫天竺鼠後,各疫苗對肺泡巨噬細胞數目及吞噬放 線桿菌之影響,以瞭解這些抗原中,何者影響最大,以及假性狂犬病疫苗是因病毒顆粒或是 因佐劑的作用,才引起肺泡巨噬細胞數目的增加。實驗結果,在各免疫組中,不論為那一種 抗原,均能引起肺泡巨噬巨噬細胞數目的增加。但其中以油劑假性狂犬病疫苗引起細胞增加 所需的時間最短,數目最多,維持增加的時間最長,在免疫後,每毫升肺臟沖洗液內巨噬細 數目,第一週為23.82±3.64×106個,第二週為44.96±9.60×106個,第三週為41.56±7.46×106 個,而未經免疫之對照組則為12.8±2.67×106個,故與對照組比較,分別為1.86倍,3.51 及3.24倍之增加,在吞噬細菌能力的比較方面,結果與數目增加甚為相似,所有免疫組之巨 噬細胞吞噬能力,均比照對組明顯的增加(P<0.05),而其中仍以油質假性狂犬病疫苗所引起 影響最為顯著,由本實驗之結果,清楚的證明假性狂犬病病毒顆粒本身,或是其佐劑,均能 引起肺泡巨噬細胞數目及吞噬能力的增加,而二者合併使用,其效果更佳。 |
| 英文摘要 | The alveolar macrophages play an important role in removing particles and microorganisms from the respiratory tract, and maintain the sterility of the alveolar spaces. In order to understand the effects of vaccination on alveolar macrophages and the best vaccine to activate the alveolar macrophages to phagcoyize the germ as Actinobacillus pleuropneumoniae, we compared the abilities of the alveolar macrophage activation of several vaccines and adjuvants in guinea pigs. They included the commercial oil and liquid types of pseudorabies vaccines, the bacterin of Actinobacillus pleuropneumoniae, the LPS vaccine of hog cholera, the Freund's complete and incomplete adjuvants, and the pseudorabies virus particles. The results indicated that the best alveolar macrophage activations were found in the oil type pseudorabies vaccine which increased more significant in number of the lavaged cells than other types of vaccines. The effects of two adjuvant types of pseudorabies vaccines were similar, but the oil type was more sensitive in the level of stimulation. If the animals vaccinated the antigens without adjuvants or the adjuvants alone, although the evidence of activation was obtained, the macrophage activaties were less in intensity. Conclusively, the alveolar macrophages could be activated to enhance their microbicidal abilities after different kinds of vaccination particularly the pseudorabies vaccines. Both antigen and adjuvant also contributed their influcence on the macrophage activities for the purpose of disease prevention in the lung. |
