中文摘要 | 本研究目的在於探討相思樹根瘤菌之世代時間及其生長之生理特性等 基本資料。分離自臺灣相思樹(代號AC)、取莢相思樹(AA)、直幹相思樹 (AM) 根瘤菌共6株之9項生理生化特性,包括世代時間、培養液酸鹼度之變化、菌落 大小、產酸或產鹼反應、黏質物之產生以及在39℃、不同pH值及 2%氯化鈉之 耐受性、剛果紅之吸收與四種有機鹽類對其生長之影響等等。經由菌落計數法測 定世代時間,結果顯示,菌株編號AM14、AC19、AA20、 AM23為快生型菌株, 其世代時間皆低於2小時,AA2、AC30為慢生型菌株,其世代時間高於6小時。 產酸或產鹼反應經由溴麝香草酚藍洋菜培養基培養後之顏色變化,菌株編號 AC19、AA20、AM23產酸,而AM14、AA2、AC30 則產鹼。在洋菜YEM培養 基培養2-7天後,測其菌落大小,結果產酸之根瘤菌其皆大於1mm,而產鹼者 皆小於1mm。由此可知產酸型根瘤菌其菌落直徑一般大於1mm,且為快生型。 而產鹼型根瘤菌其菌落直徑一般小於1mm,但其可能為快生或慢生型。剛果紅 之吸收特性為相思樹根瘤菌與其它如大豆、山黃麻屬、銀合歡、花生等根瘤菌間 最明顯之差異,編號 AM14、AC19 、AA20、AM23可以吸收剛果紅,AA2、 AA30則否。大部分菌株可以在pH值介於4.0至9.5之間生長,在pH值3.5則 否。在RBM培養基分別添加20mM檸檬酸鹽、乳酸鹽、乙醛酸鹽會抑制菌株生 長,而丙酮酸鹽則會促進之。 |
英文摘要 | The physiological characteristics for Rhizobium strains isolated from Acacia confusa (AC), Acacia auriculiformis (AA) and Acacia mangium (AM) were investigated. The objectives were to determine the nine physiological characteristics of six Rhizobium strains including generation time, the ability to absorb congo red, the ability to produce acids or alkaline, mucoid production, growth in 2% NaC1, growth at 39℃, growth at different pH and the ability to grow on basal salts medium containing sucrose and arabinose as well as the presence of 20 mM citrate, pyruvate, lactate or glyoxylate. Results showed that the generation times of AM14, AC19, AA20 and AM23 estimated by colony count were under 2 hours and were regarded as the fast-growing strains. The generation time of AA2 and AC30 exceeded 6 hours and were classifried as the slow-growing strains. Incubation of AC19, AA20, and AM23 with 0.5 (v/v) bromthymol blue on yeast-mannitol-mineral (YEM) salts agar and liquid YEM medium resulted in acid production. The same treatment yielded alkaline reaction for AM14, AA2 and AC30. Colony diameter were measured after 2-7 days of culturing on YEM salts agar. Acid-producing rhizobium were measured to be greater than 1 mm when alkaline-producing rhizobium were less than 1 mm. Thus, the colony size of the acid-producing rhizobium strains used in this study is always greater than 1 mm and all belong to the fast growing strains. Consequently, the colony size of the alkaline-producing rhizobium strains is constantly under 1 mm but may either be fast or slow-growing strains. Strains AM14, AC19, AA20 and AM23 have the ability to absorb congo red from a YEM salts medium. This characteristics is unique among rhizobia isolated from soybean, Trema cannabina Lour., Leucaena spp., and peanut. Most strains can grow within the pH range of 4.0-9.5 the but growth is restricted at pH 3.5. The rhizobium basal medium (RBM) containing 20 mM of citrate, lactate or glyoxylate inhibited bacteria growth where as pyruvate stimulated bacteria growth. |
