中文摘要 | 分析複雜的血清蛋白變化對燒傷病人的處理是很重要的。本研究嘗試以只需要少量血清,可同時定性與定量分析多種蛋白成分變化的雙向電泳進行分析。結果顯示與正常對照組血清相比較燒傷病人血清中antitrypsin, haptoglobin, albumin, transferrin, immunoglobulins,及hemoglobin含量均有變化。此外,有三種蛋白成分僅在燒傷病人血清中被發現。與己報告之文獻相比對,其中分子量約42000道爾頓,pI值接近類似免疫球蛋白IgG 重鍵的血清蛋白成分為creatine kinase (CK)。其 它兩種蛋白成分,以特異性抗血清及免疫轉印法證實為C-reactive protein (CRP)及myoglobin。CRP在本研究所使用的二位燒傷病人血清中均可測得,而CK及myoglobin則僅在其中一位已死亡燒傷病人的血清中被發現。此外,在此病人燒傷後第十天,同時也是在病人死亡前二十四小時內所收集的血清內,尚發現有分子量介於64000與36000道爾頓,pI值介於antitrypsin與IgG重鍵問之不正常血清蛋白成分存在。這些結果所代表的意義將在文中進行討論。而本實驗結果亦顯示雙向電泳是分析燒傷病人血清蛋白變化的良好工具。 |
英文摘要 | It is important for clinical management of burn patients to analyze complex changes in serum proteins. The present study showed that serum protein changes in response to burn injury can easily be analyzed by twodimensional (2D) gel electrophoresis which allows simultaneous analysis, both qualitatively and quantitatively, of numerous proteins from smallvolume samples. Changes in concentrations of such proteins as antitrypsin, haptoglobin, albumin, transferrin, immunoglobulins, and hemoglobin in sera from burn patients were compared with those from normal controls, and were identified. Furthermore, three components which appeared only in 2D gels of post-burn sera were recognized. The component with a molecular weight (MW) of about 42,000 daltons and a pI value similar to that of the heavy chains of IgG was identified as creatine kinase (CK) by comparison with 2D maps reported previously. The other two components were identified as C-reactive protein (CRP) and myoglobin, respectively, by immunoblotting using specific antisera. Among these three components, CRP has been identified in sera from two patients included in this study. However, both CK and myoglobin were identified only in sera from a patient who had expired, but not in sera from the surviving patient. Furthermore, abnormal protein constituents with molecular weights between about 64,000 and 36,000 daltons and pI values between those of antitrypsin and heavy chains of IgG were also observed in a Day 10, post-burn serum which was collected within 24 hours before death. The significance of these findings is discussed. Indications, based on this study, are that 2D gel electrophoresis can be a valuable tool in analysis of sera from burn patients. |
