| 中文摘要 | 本文報告測定人循環血液中血清免疫複合物(CIC)的一種敏感而簡便的改良方法,此法係利用Clq與酵素結合形成免疫複合物(ELISA)的原理來測定。因酵素安全而穩定,故以此做反應標記遠較以放射性同位素(superscript 125)I為佳。此法僅需微量之試劑,故又經濟,而且操作簡便,利用Titertek multiskan可半自動化地讀記微板孔內酵素反應之顏色強度,以定性或定量CIC,於1分鐘內得到結果。此法重複性(reproducibility)甚高,經測定在相當0.5至3µg凝集人類免疫球蛋白G(AHG)之範圍內非常敏感,與其它方法相當或更佳。 經對30名正常人及83例患者;其中包括系統性紅斑性狼瘡(SLE)23例,類風濕性關節炎(RA)25例,惡性腫瘤(MT)(肺癌10例,肝癌9例)19例,和急性B型肝炎(HB)16例;以此改良之測定方法進行血清中CIC的測定,其平均值(以µg AHG eq/ml表示)在正常對照組為13.7±6.12,SLE為67.93±42.22,MT為41.66±24.6,RA為29.54±15.24,HB為27.34±17.79,以上四組患者之平均值與正常對照組相比皆有顯著之差異(P<0.001)。此結果與其他學者報告用其它方法相比甚為相近或更佳。因之,此微板酵素結合免疫吸附法係一安全、簡便、經濟和敏感之血清免疫複合物測定之方法。 |
| 英文摘要 | A sensitive and simple procedure for detection and quantitation of circulating immune complexes (CIC) in human sera has been developed by utilizing Clq-microplate ELISA system. The utilization of an enzyme marker reagents, as opposed to 12S I-labled reagents is extremely advantageous, since it is safe, stable, economical, and fast. We applied this system to detect CIC in sera of 30 normal healthy controls and 83 patients with various diseases, including 23 cases of systemic lupus erythematosus (SLE), 25 cases of rtheumatoid arthritis (RA), 19 cases of malignant tumor (MT) (pulmonary carcinoma in 10 and hepatoma in 9 and 16 cases of acute type B hepatitis (HB). The average level of CIC (ug AHG eq/ml) in normal control group and four different patient groups were as follows: 12.7±6.l2 for normal group, 29.54±15.24 for RA, 67.93±42.22 for SLE, 41.66±24.6 for MT and 27.34±17.79 for HB. The average of all the four different patient groups was significantly higher than that of the normal control group (P<0.001). Our results were some what better or equivalent tothat by other methods in the literature. The conclusion that C1q microplate ELISA system is a safe, simple, economical and sensitive method for detecting circulating immune complexes is thus made. |
