中文摘要 | (一) 利用Blaydes培養基供給各2 ppm的 2, 4-D及Kinetin,可成功的誘導甘藷花葯形成Callus, IAA對Callus之誘導液沒影響。提高2, 4-D及Kinetin之濃度可抑制Callus形成後之繼續生成。 (二) 不同品種間,Callus誘導之成功百分比各有差異。 (三) Callus 的顏色和各品種之塊根肉色一致。 (四) Callus 的繼代培養,在IAA, 2, 4-D及Kinetin三種auxins中, 2, 4-D最為重要,若以各2 ppm的 2, 4-D與IAA配合使用,則Callus之生長情形更為良好。 (五)數種受測之植物抽出液中,碗豆抽出液Callus對生長促進之效果最好,特別是和2, 4-D配合使用時,更可提高其促進效果;椰子胚乳液單獨使用效果極低,但若和2, 4-D配合則效果大大增加。Callus生長最良好之培養基為含0.5-3 ppm 2, 4-D, 15%椰子胚乳液及10%碗豆抽出液之Blaydes培養基。 |
英文摘要 | After inducing the callus formation from in-vitro cultured sweet potato anther, the primary objective of this study is to find out the most suitable culture medium for the rapid increase of the induced callus with a short period. The effects of IAA, 2, 4-D and Kinetin as well as several plant extracts were carefully studied. The results obtained from these studies are summarized as follows: (1) Callus formation from anther was studied by inoculating the anther into the Blaydes' medium supplemented with 2 ppm., each of 2, 4-D and Kinetin. IAA was found to have little effect on callus induction. Higher concentration of 2, 4-D and Kinetin appeared to suppress the subsequent growth of callus thus induced. (2) The percentages of the callus-forming anthers against the total numbers of inoculated anthers were different among differernt varieties. (3) The color of the induced callus was same as that of the flesh of root tuber of respective varieties. (4) For subculturing the induced callus, 2, 4-D was most influential among 2, 4-D, IAA and Kinetin. Best growth of callus in subculture was obtained with medium supplemented with 2 ppm., each of 2, 4-D and IAA. (5) Of the several plant extracts studied, pea extract was found most effective in increasing the growth of callus, particularly when combined with 2, 4-D. Coconut milk alone showed very little effect on the growth of callus, but when combined with 2, 4-D the effect would be greatly enhanced. Extremely vigorous callus growth was obtained from the medium supplemented with 0.5-3 ppm. 2, 4-D, 15% of coconut milk and 10% of pea extract. |
