中文摘要 | (一)不同品種的甘藷花葯Callus,對IAA, 2, 4-D及Kinetin等3種auxins的反應各有不同;新竹1號Callus對2, 4-D的反應比臺農59號Callus強,但對IAA及Kinetin之反應,則為臺農59號比新竹1號強。 (二)一般無機鹽類或維他命對Callus生長之效果,沒有IAA, 2, 4-D及Kinetin等3種auxins來得顯著。各種有機,無機鹽類及auxins之適當濃度,以新竹1號及臺農59號2品種Callus,利用三角形法分別測定,所得結果如表7所列。 (三)培養基中Sucrose之濃度對Callus生長之影響極大,最適濃度為4~5%,將Sucrose濃度提高至7%以上時,Callus生長會被抑制,但Callus之乾物量卻隨著增加。 (四)Callus經繼代培養後,最初1個月生長期間,pH值可影響Callus之生長,最適pH為6.25~6.5間;若培養期限延至2個月時,所得最初不同之pH值,均穩定於pH=5.15~5.35間,且pH=5~7之各處理,Callus生長並無差異。 (五)甘藷花葯Callus生長之最適溫度為30℃。 (六)黑暗下生長之Callus,比人為照光好。 (七)15 mg Callus經30天生長,改良培養基優於BLAYDES培養基之百分比,臺農59號Callus為765%,新竹1號Callus為3,084%。 |
英文摘要 | After inducing the callus formation from in-vitro cultured sweet potato anther, the primary objective of this study is to find out the most suitable culture medium for the rapid increase of the induced callus within a short period. Each composition included in the Blaydes' medium was carefully studied and evaluated. And the effects of sucrose concentration as well as the cultural conditions on the growth of callus were also studied. The results obtained from these studies are summarized as follows: 1. Varietal differences were noticed in the response to different auxins. The variety Hsinchu No. 1 responded more strongly to 2, 4-D, but less to IAA and Kinetin than Tainung No. 59. 2. In general, the effects of inorgani csalts and vitamins on the growth of callus were not so significant as those of auxins. By applying the three-salt trianguar method, the optimum concentrations of various inorganic salts and vitamins were determined for the Hsinchu No. 1 and Tainung No. 59 (as shown on Teble-7). 3. The conentration of sucrose in culture medium showed a highly significant influence on the growth of callus. The optiman concentration was found within the range of 4-5%. Higher conentration than 7% caused growth retardation, though the percentage of dry weight of callus to the fresh weight were much higher when sucrose concentration was higher. 4. When the subculture duration was limited to 30 days. the pH value of the culture medium affected greatly the growth of callus. The optimum pH value lay between 6.25 and 6.50. However, when the culture duration was prolonged to 60 days, all different pH values of initial cultural media became stabilized at the range of 5.1-5.3 and no difference in callus growth was found among culture media having pH value between 5 to 7. 5. The optimum temperature for incubating the subculture callus was found to be 30℃. 6. Callus subculture in darkness showed much larger growth than that exposed to light. 7. Callus of Tainung No. 59 and Hsinchu No. 1, when grown by the improved medium under improved cultural conditions thus worked out in this study, outyielded those grown with Blaydes' original medium by 765% and 3,084% respectively after 30 days of subculture. |
