| 中文摘要 | 本研究自田間採集並篩選對桃蚜具致病潛力之蟲生真菌,再進行此菌株培養特性之探討。自不同區域土壤中分離出黑殭菌(Metarhizium anisopliae)、球孢白殭菌(Beauveria bassiana)、淡紫菌(Purpureocillium lilacinum)及Pochonia chlamydosporia等共11種菌株。將各分離菌株孢子懸浮液濃度配製成10^7 conidia/ml,於室內25℃及高相對濕度環境下進行對桃蚜成蟲之致病力篩選,初步顯示黑殭菌CCC 901菌株於接種7天後可達較佳致死效果,平均死亡率可達86.7±1.7%。進一步試驗CCC 901之不同濃度孢子懸浮液(10^5、10^6、10^7及10^8 conidia/ml)分別對桃蚜成蟲及二齡若蟲之致病力,結果顯示CCC 901菌株對桃蚜成蟲及二齡若蟲之半數致死時間(LT_(50)),均以孢子濃度10^8 conidia/ml時最短,分別為2.3及2.6天。模擬設施環境(77% RH)內評估CCC 901對桃蚜之防治效率,分別以CCC 901以10^6、10^7及10^8 conidia/ml濃度,間隔10天連續2次施用後,結果顯示至第10日之平均校正防治率分別達51.1、73.7及78.9%。另以菌株培養時的菌落直徑及產孢量為評量依據,探討最適合CCC 901菌株生長之溫度及光週期等,結果顯示此菌株於Sabouraud Maltose Agar Yeast(SMAY)平板上,適合培養於25℃且全光照之環境條件中。另試驗化學農藥對此菌株之影響,顯示亞托敏(azoxystrobin)23% SC會抑其生長及產孢,而固殺草(glufosinate-ammonium)18.02% SL雖未發現抑制菌株生長,但會影響其產孢量。相關試驗結果顯示此菌株具潛力開發為管理桃蚜之微生物製劑。 |
| 英文摘要 | In this experiment, the native entomopathogenic fungi of Myzus persicae were collected and screened from the field, and the pathogenicity test and culture characteristics were discussed. A total of 11 isolates, including Metarhizium anisopliae , Beauveria bassiana , Purpureocillium lilacinum and Pochonia chlamydosporia , were isolated from the soil of different regions. The pathogenicity of the adults of Myzus persicae was conducted by using the conidial suspension of each isolate (10^7 conidia/ml) as the inoculum under high relative humidity in the laboratory (25℃). The preliminary result showed that the isolate of Metarhizium anisopliae CCC 901 with better lethal effect after 7 days of inoculation, and the average mortality was 86.7±1.7%. Further, the pathogenicity of CCC 901 conidial suspensions with different concentrations (10^5, 10^6, 10^7 and 10^8 conidia/ml) inoculated on adults and 2^(nd) instar nymphs of Myzus persicae were tested. The results showed the less LT_(50) values of CCC 901 (10^8 conidia/ml) against adults and 2^(nd) instar nymphs were 2.3 and 2.6 days, respectively. Evaluate the control efficiency of three concentrations of the CCC 901 conidial suspensions (10^6, 10^7 and 10^8 conidia/ml) in a simulated facility environment (77% RH) against Myzus persicae. The results showed that the concentrations of CCC 901 of 10^6, 10^7 and 10^8 conidia/ml, after two consecutive applications at intervals of 10 days, the average corrected control efficacy up to the 10th day were 51.1, 73.7 and 78.9%, respectively. Based on the colony diameter and sporulation, the most suitable temperature, photoperiod and compatibility with pesticides for the growth of CCC 901 were tested. The results showed that it was the most optimal to be cultured on Sabouraud Maltose Agar Yeast (SMAY) medium at 25℃ and 24 hour light cycle. Besides, azoxystrobin 23% SC inhibited the growth and sporulation of CCC 901, while glufosinate-ammonium 18.02% SL inhibited the sporulation of the isolate, with no effect on the growth of it. Therefore, M. anisopliae CCC 901 possessed the potential to be developed as microbial formulation for the control of Myzus persicae. |
