| 中文摘要 | 目的:台灣牛樟芝(Antrodia camphorata)為台灣特有種菇蕈類,大多於臺灣民間偏方及療法中使用,樟芝萃取物所含的活性物質及其代謝物已證實具有多種藥理活性,在動物實驗中亦證實具有包括清除活他態的致癌劑或致突變劑、調節細胞週期蛋白等,然而對攝護腺癌細胞的抑制效果則未曾有系統的研究;攝護腺癌發生率近年來有顯著上升的趨勢,因此如何預防與治療攝護腺癌已成為重要的課題。方法:本研究以LNCaP cell line(human prostate adenocarcinoma cells;人類攝護腺癌細胞株)進行體外培養,探討樟芝萃取物對於攝護腺癌細胞凋亡(apoptosis)訊息路徑的影響, 我們以流式細胞儀(flow cytometry)分析癌細胞之細胞週期、細胞凋亡以及使用西方墨點法分析細胞凋亡相關訊息路徑變他情形與下游蛋白質的表現,以進一步探討樟芝萃取物抑制攝護腺癌細胞之機轉。結果:研究結果顯示,細胞在加入不同濃度之樟芝萃取物後,經流式細胞儀(flowcytometry)分析後發現樟芝萃取物明顯誘發細胞凋亡反應,顯示樟芝萃取物確實具有促使癌細胞細胞凋亡之能力,然後經西方墨點法分析後發現,樟芝萃取物亦明顯抑制Bc1-2蛋白進一步誘發細胞凋亡蛋白Bax、Cyt-C、Caspase 8、Caspase9、Parp,研究結果顯示樟芝萃取物藉由誘發細胞凋亡蛋白之作用機轉來達成抑制癌細胞之效果。結論:因此本研究成功證實樟芝萃取物具有抗攝護腺癌細胞之能力與其可能之抗癌作用機轉,本研究不止促進抗癌細胞相關機制的瞭解,亦可開發出全新抗癌之替代療法可作為臨床醫師之參考,希望可以大大降低本國花費於攝護腺癌上的醫療社會成本。 |
| 英文摘要 | Objective: Antrodia camphorata extracts is an important traditional Chinese medicine for the treatment of human diseases such as food and drug intoxication, diarrhea, abdominal pain, hypertension, itchy skin and liver cancer. The potential probability of anti-oxidant and anti-mutagenesis of Antrodia camphorata were well studied. The anti-cancer by Antrodia camphorata extracts against prostate cancer were never been systematic evaluated. The purpose of this study is to verify the inhibitory effect on prostate carcinogenicity by Antrodia camphorata extracts. Methods: The evaluation methods including modulation of signal transduction of apoptosis, shift of cell cycle by flow cytometry, and apoptosis proteins analysis by Western blots. The mechanisms of anti-carcinogenesis, evaluation models and new function-potential screening standard process will be established on Antrodia camphorata extracts in this study. Results: The results indicated the amounts of Bax, Cyt-C, Caspase8, Caspase9, Parp and the apoptosis effects were dose-related induced. The results of this study can provide the experimental evidence that inhibitory effect on anti-carcinogenicity of prostate tumors by Antrodia camphorata extracts. Conclusions: The potential benefits of anti-cancer drugs will be promoted and enhances the international competitiveness largely and will been proven to be helpful for understanding the anti-carcinogenesis mechanism by Antrodia camphorata extracts and provide novel insights in controlling prostate cancer in our population. |
