中文摘要 | 中藥萃取物所舍的活性物質及其代謝物已證實具有多種藥理活性,在動物質驗中亦證實具有抑制腫府生長的作用,研究指出山芙蓉(Hibiscus Taiwanensis, HT)具有包括清除活性氧自由基、提高免疫力等;然而對大腸癌細胞的抑制效果則未曾有系統的研究,臨床上大腸癌的發生率近年來有顯著上升的趨勢,因此如何預防與治療大腸癌已成為重要的課題。方法:本研究以HT29 cell line(human colorectal adenocarcinoma cell line;人類結腸直腸腺癌細胞株)進行體外培養,探討山芙蓉萃取物對於大腸癌細胞凋亡(apoptosis)訊息路徑的影響,我們以流式細胞儀分析癌細胞之細胞週期、細胞凋亡以及使用西方墨點法分析細胞凋亡相關訊息路徑變他情形與下游蛋白質的表現,以進一步探討山芙蓉萃取物抑制大腸癌細胞之機轉。結果:細胞再加入不同濃度之山芙蓉萃取物後,經flow cytometry分析後發現心芙蓉萃取物明顯誘發細胞凋亡反應,顯示山芙蓉萃取物時實具有促使癌細胞細胞凋亡之能力,然後經西方墨點法分析後發現,山芙蓉萃取物亦明顯誘發細胞凋亡蛋白caspase3/7、PARP(Poly(ADP-ribose)polymerase)等並明顯抑制細胞凋亡抑制蛋白BCL-xL,本研究結果證實山芙蓉萃取物可透過誘發細胞凋亡蛋白質之含量,並透過抑制細胞凋亡抑制蛋白質之含量以減少對;凋亡蛋白的抑制作用來達到誘發細胞凋亡反應的效果。結論:本研究成功證實山芙蓉萃取物具有抗大腸癌細胞之能力與其可能之抗癌作用機轉,本研究不止促進抗癌細胞相關機制的瞭解,亦可開發另一種抗癌之輔助療法。 |
英文摘要 | Hibiscus Taiwanensis extracts were proved be capable of free radicals cleavage and enhancement on human immunity. The anti-cancer effects by Hibiscus Taiwanensis extracts against colon cancer were never been systemic evaluated. The purpose of this study is to verify the inhibitory effect on colon carcinogenicity by Hibiscus Taiwanensis extracts. Methods: In this study, we used Hibiscus Taiwanensis extracts to inhibit colon cancer cell proliferation and tumor growth, and investigate the underlying molecular mechanisms. HT-29 cell line ( human colorectal adenocarcinoma cell line) was used in this study. Cell cycles were analyzed by Flow cytometry. Signal transductions were analyzed by western blots. Results: The results indicated the amounts of caspase3/7 and parp (Poly (ADP-ribose) polymerase) and the apoptosis effects were dose-related induced. The amounts of BCL-xL, the inhibitor of apoptosis, were dose-related decreased. Hibiscus Taiwanensis extracts were proved be capable of modulation of apoptosis via apoptotic protein induction and antiapoptotic protein deduction. Conclusions: The results of present study can provide the experimental evidence that inhibitory effect on anticarcinogenicity of colorectal adenocarcinoma by Hibiscus Taiwanensis extracts. |
