中文摘要 | 研究指出,何首烏( Polygonum multiflorum)具有多方面的療效,如抗氧化、抗高血脂、調節膽固醇的吸收,因此可防止血管硬化;其次增強免疫力、延緩老化、保肝作用、抗菌能力。由於現代人對於抗老化食品、藥物的訴求日漸增高,因此擁有多種療效的何首烏被廣泛使用。本研究目的為探討何首烏單方並加入兒茶素作複方對於抗老化、促進小鼠學習記憶、行為能力、體內抗氧化狀態、腎功能及血脂之影響。本實驗使用由國家動物中心購買之 6周齡 C57BL/6雌性小鼠,首先我們設計在小鼠雙側進行摘除卵巢手術,休養後皮下注射 D-半乳糖(D-galactose,DG),為期 4週後建立老化小鼠模型。實驗分成 5組:控制組(Normal)、負對照組( Saline)、實驗組(何首烏單方)、對照組(兒茶素單方)、實驗組(何首烏 +兒茶素)餵食 500mg/kg劑量,為期 4週。實驗過程期間記錄小鼠體重變化及老化指數評估,如行為方面進行活動量試驗,並以單次被動迴避試驗及主動迴避試驗進行學習記憶能力測試。同時實驗期間也進行血液生化分析,觀察腎功能指數之變化,肌酸酐( Creatinine,Cre.)、尿氮素( BUN)、 Albumin-Creatinine Ration(ACR)試驗,並進行 lipid profile試驗(檢測總膽固醇(Total Cholesterol,TC)、三酸甘油酯( Triglyceride,TG)、高密度脂蛋白( High Density Lipoprotein,HDL)、低密度脂蛋白( Low Density Lipoprotein,LDL)的含量,觀察血脂代謝是否因老化而異常。評估給藥前後對於老化動物模式,是否有改善血液相關生化值之功效。實驗結果顯示 : 在老化指數評估方面,實驗組(何首烏 +兒茶素)之老化指數總分低於對照組。而行為活動量試驗方面,經主動迴避試驗結果發現實驗組(何首烏 +兒茶素)均有較佳的學習記憶能力;經單次被動迴避試驗發現實驗組(何首烏 +兒茶素)在學習記憶保留上顯著較佳。綜合上述結果,何首烏單 /複方皆可降低血清中 TC、TG、HDL/LDL、Creatinine、 BUN與 ACR,有效改善老化動物情形。此外,在行為方面進行活動量試驗法,發現何首烏單 /複方均可藉由記憶力測試的規避次數有明顯上升,顯示能夠改善老化小鼠之學習記憶能力得以達到改善老化之功效。 |
英文摘要 | According to Traditional Chinese medicine (TCM) studies, Polygonum multiflorum (PM) has many effect as antioxidant, antihyperlipidemic and reduced cholesterol, therefore can be alleviate vascular stiffness. Secondly enhance immunity, defer aging, hepatoprotective effect, antibacterial ability. A multifunctional PM which is widely used due to high demanding of anti-aging food and medicine by modernist. This study was aimed to explore the promotion of single PM and added catechins as Compound of defer aging, advance the memory, behavior capacity, in vivo antioxidant status, affect renal and lipid profile the aged mice study. The 6-week-old C57BL/6 female mice were obtained from the National Animal Center, take rest after removed the ovary then D-galactose(DG) was injected subcutaneously for 4 weeks. Experiments were divided into five groups: control group (Normal), the negative control group (Saline), the experimental groups (single PM), the control group (catechin), the experimental groups (PM + catechin) at a dose of 500mg/kg, for a period of four weeks. During the course of the experiment record weight changes and aging index assessment such as behavioral aspects of the activity test, a single trial passive avoidance, active avoidance test learning and memory tests. Also conducted blood biochemical analysis during the experiment, observe whether renal function index rises due to aging (Creatinine, Blood Urea Nitrogen, Albumin-Creatinine Ration test) and conducted lipid profile test (total cholesterol, triglyceride, high-density lipoprotein, Low-density lipoprotein)for observe whether lipid profile index reduce due to aging and abnormality. Assessment the relevant biochemical values of the effect before and after administration of aging animal model. The results show the aging evaluation: experimental groups (PM + catechin) aging index Total than the negative control group (Saline). Behavioral aspects of the activity test, experimental groups (PM + catechin) have better learning-memory; in a single trial passive avoidance and active avoidance test, experimental groups (PM + catechin) have better learning-memory. In conclusion that the PM single and compound has reduce the TC, TG, HDL/LDL, Creatinine, BUN and ACR ratio. Moreover a behavioral aspects of the activity test shows PM single and compound has index rises learning-memory, improve the relevant values of an aged mice models for aging effect. |
