中文摘要 | 本研究中自野外純化分離出兩株產油藻株並以18S rDNA分子鑑定命名為Chlorella sorokiniana CPC3-1及Ettlia texensis CPC5-1。探討混營培養時碳源及氮源對微藻生產及產油率之影響,結果以Crude Glycerol及Glycine對CPC3-1較佳;Glucose及Monosodium Glutamate(MSG)對CPC5-1為較佳之碳源及氮源。混營培養條件經過最適化探討後,CPC3-1生物質量可達1.977 ± 0.016 g/L,粗脂肪含量為48~62%;CPC5-1藻體乾重可達1.727 ± 0.027 g/L,粗脂肪含量為46~52 %。分析CPC3-1藻油脂肪酸組成以Palmitic Acid(C16:0)最多,其次是Linoleic Acid(C18:2)及Myristic Acid(C14:0);CPC5-1藻油脂肪酸組成以C16:0為最高,其次是C16:2及C14:0。這兩個新藻株未來應用於商業以生物反應器混營產油上頗具潛力。 |
英文摘要 | This article will be discussing the two novel green microalga obtained from local freshwater in Taiwan, also named as "Chlorella sorokiniana" CPC3-1 and "Ettlia texensis" CPC5-1, identified by 18S rDNA molecular typing. Additionally, this research will be evaluating the effects of mixotrophic cultivations on biomass and lipid production. By searching the preferences of the carbon and nitrogen sources has revealed that these strain could grow photoheterotrophically well with crude glycerol and glycine for CPC3-1, and glucose and monosodium glutamate (MSG) for CPC5-1. In the optimized medium, the highest biomass concentration and total crude lipid content achieved were 1.977 ± 0.016 g/L and 48~62% of dry weight basis of CPC3-1; 1.727 ± 0.027 g/L and 46~52% of dry weight basis of CPC5-1. Moreover, the major fatty acid methyl esters were palmitic acid (C16:0), linoleic acid (C18:2) and myristic acid (C14:0) of CPC3-1; C16:0, C16:2 and C14:0 of CPC5-1. These two new isolates could be a potential candidate for the commercial mixotrophic production in the bioreactor. |
