中文摘要 | 本論文探討綠豆癀對大白鼠之急性毒性評估,採用Litchfield and Wilcoxo方法,分正常組及實驗組(綠豆癀粉末劑量:0.0 mg/kg、1,000 mg/kg、3,000 mg/kg、10,000 mg/kg),總共四組。進行口服單劑量之短期試驗,觀察大白鼠死亡數目及中毒情形以計算急性毒性之半數致死劑量(LD50)。此外,觀察期結束後採取小鼠的血液及臟器,分別檢測血清生化指標及組織病理切片。結果:急性毒性試驗中,四組皆無死亡,其半數致死量為每公斤大於10.0公克,顯示綠豆癀粉末毒性甚小。時間效應試驗中,大白鼠口服投與綠豆癀粉末(3,000 mg/kg),在不同時點(6h、24h、72h、168h)觀察其行為、生理生化及組織型態學等之影響。結果顯示,單劑量綠豆癀並無明顯毒性表現;另外卻顯示對正常老鼠之血脂、血糖有良性調控作用。 |
英文摘要 | The evaluation of acute toxicity of "Praeparatum Mungo." on rat was based on the Litchfield and Wilcoxon method. All rats were divided into four groups, one control group and three experimental groups (dose: 0.0 mg/kg, 1,000 mg/kg, 3,000 mg/kg, 10,000 mg/kg). Each dose of the powder of "Praeparatum Mungo." and normal saline was administered orally to those rats. All animals were observed 72 hours successively to evaluate their toxicological responses and mortality and thus ca1culate the semi-lethal dose (LD50). After 72 hours, al1 animals were sacrificed and their blood and target organs were collected. The collected sera were examined to determine the biochemical function, inc1uding liver function (GOT, GPT), nephritic functions(BUN, Creatinine) and metabolic function (cholesterol, glucose). The result revealed the powder of "Praeparatum Mungo" is extremely slightly toxic (LD50>10.0 gm/kg). In time course test the powder of "Praeparatum Mungo" was administered orally to rats. Then in every interval of 6, 24,72 and 168 hours, the experimented animals were observed and evaluated according to behavior, biochemical index and histopathological observation. The results showed that non particular toxicity of "Praeparatum Mungo" was found. In the meanwhile it showed positive functions of modulation in serum lipid file and glucose on rats. |
