中文摘要 | 本實驗使用1.5月齡年輕大鼠及15月齡中年大鼠,探討人尿製備連續八週經口投與的毒性作用。年輕大鼠投與劑量分別為0.3,1.0,3.0 g/kg,中年大鼠投與劑量分別為 0.3,1.0 g/kg。血液學檢查:年輕大鼠3.0 g/kg組及中年大鼠1.0 g/kg出現血小板增加症,中年大鼠0.3、1.0 g/kg組出現淋巴球比率增加及中性球比率下降的情形,中年大鼠1.0 g/kg也出現部分凝血活�t時間減短作用。血清生化學檢查:年輕大鼠3.0 g/kg組及中年大鼠1.0 g/kg出現血糖上升情形,年輕大鼠3.0 g/kg組鉀離子減少,中年大鼠1.0 g/kg組氯離子下降,中年大鼠0.3 g/kg及1.0 g/kg組的尿酸減少。尿液分析顯示;年輕大鼠3.0 g/kg組及中年大鼠1.0 g/kg組尿中鈉離子增加,年輕大鼠3.0 g/kg組及中年大鼠0.3、1.0 g/kg組尿蛋白隻數較多。中年大鼠1.0 g/kg組的前列腺重量增加。結論:以年輕大鼠而言在1.0 g/kg劑量下沒有毒性出現。 |
英文摘要 | The 8-week repeated oral dose toxicity study of a preparation of human urine (PHU) was evaluated in both male young rats (1.5 months) and middle-aged rats (15 months). PHU was administrated orally at once-daily doses of 0.3, 1.0, 3.0 g/kg in young rats and 0.3, 1.0 g/kg in middle-aged rats. In hematology, young rats given 3 g/kg and middle-aged rats given 1 mg/kjg showed thrombocytosis. Increased percentages of lymphocyte and decreased percentages of segmented neutrophil were observed in middle-aged rats of the 0.3 and 1.0 g/kg dose groups. In addition, a decreased activated partial thromboplastin time was observed in middle-aged rats of the 1.0 g/kg dose group. In blood chemistry, an increased glucose level was observed in young rats of the 3 g/kg dose group and in middle-aged rats of the 1 g/kg dose group. Young rats given 3 g/kg showed a decrease in K�� level. Middle-aged rats given 1.0 g/kg showed a deceased in Cl�� level. A decreased uric acid level was observed in middle-aged rats of the 0.3 and 1.0 g/kg dose groups. Urinalysis showed an increase in Na�� level in both young rats of 1.0 g/kg dose group. Increased urinary protein was observed in young rats of the 3.0 g/kg dose group and middle-aged rats of 3.0 g/kg dose group and middle-aged rats of 1.0 g/kg dose group. Increased urinary protein was observed in young rats of the 3.0 g/kg dose group and middle-aged rats of 0.3, 1.0 g/kg dose groups. Middle-aged rats given 1.0 g/kg showed an increase in prostate wight. In conclusion, the toxicological no-observed levels in young rats were thought to be 1.0 g/kg or below. |
