中文摘要 | 為探討臺灣產五種魟類(黑土魟Dasyatis navarrae、古氏土魟D. kuhlii、廣東老板鯆Dipturus kwangtungensis、雪花鴨嘴燕魟Aetobatus narinari、和中國黃點鋪Platyrhina sinensis)之粒線體細胞色素b基因之完整1,141 bp核酸序列,使用4組引子(L14735/H15149ad、L14734-Glu/H15557-CYB、CB4aL-15040/CB6ThrH-15930、和L15460/CB6ThrH-15930)進行聚合酶鏈鎖反應增幅,得知五種魟類之核酸變異位數有164-297個,其中Di. kwangtungensis和P. sinensis皆屬於鯆目Rajifomes,但細胞色素b之核酸序組成與燕魟目Myliobatiformes者差異不大。其中五種魟類除A. narinari外,其餘四種魟類之粒線體細胞色素b基因序列均為首次報導。依此基因序列推算五種魟類之種屬相關性,則與傳統型態學分類者相同。 |
英文摘要 | In order to determine the complete 1,141 bp nucleotide sequence of mitochondrial cytochrome b gene for five common species of stingray in Taiwan (including Dasyatis navarrae, D. kuhlii, Dipturus kwangtungensis, Aetobatus narinari and Platyrhina sinensis), four sets (L14735/H15149ad, L14734-Glu/H15557-CYB, CB4aL-15040/CB6ThrH 15930 and L15460/CB6ThrH-15930) of fish-versatile primer were used to amplify three continuous and overlapping entire cytochrome b gene sequences. The complete 1,141 bp cytochrome b gene sequences have for the first time been applied to calculate base compositions and genetic distances. The genetic distances calculated from cytochrome b genes of five common species of stingray in Taiwan indicate relationships between species to similar to those already shown by studies using the traditional morphological taxonomic approach. |
