中文摘要 | 本 研 究 之 目 的 在 嘗 試 以 高 效 液 相 層 析 法 開 發 同 時 檢 測 雞肉及豬肉中八種 quinolone 類抗菌劑 (oxolinicacid, nalidixicacid, flurnequine, ciprofloxacin danofloxacin,enrofloxacin,ofloxacin 及 sarafloxacin) 之方法。 以 acetonitrile 及甲醇氫氧化鈉溶液二次萃取, 再利用 n-hexane 移去脂溶性物質,經減壓 濃縮後, 以 acetonitrile-O.1% triethylamine (3.5: 6.5) 1ml 超音波抽出, 再以 n-hexane 抽出脂溶性物質而後進行分析,以裝有保護管柱之分離管 (Cosmosil 5C18-MS), 配合移動相 acetonitrile-O.1% triethylamine + 7 mM sodium dodecyl sulfate (35:65)(pH 3.0),搭配紫外光瞬間變換波長 (0-11 分波長 254 nm:11-30 分波長 282 nm) 可精確地同時分析上述八種 quinolone 類抗菌劑。各組織之回收率為 73.5-90.7%。最低檢 測極限為 0.01 u g/g。 |
英文摘要 | A high performance liquid chromatographic (HPLC) method for the simultaneous determination of the residues ofquinolone drugs, including oxolinic acid, nalidixic acid, flurnequine, ciprofloxacin, danofloxacin, enrofloxacin, ofloxacin, and sarafloxacin in pork and chicken muscle was developed. The samples were prepared by extraction with acetonitrile and methanolic sodium hydroxide solution followed by liquid-liquid partition with acetonitrile saturated n-hexane for clean up. The HPLC separation was carried out on a C18 column (Cosmosil 5C18-MS column; 150 mm x 4.6 mm I.D.) with a guard column with 0.1% triethylamine buffer (pH 3.0) containing 7 mM sodium dodecyl sulfate (SDS)-acetonitrile (65:35) as the mobile phase at a flow rate of 0.7 ml/min. The chemicals were detected by UV detection (UV wavelength programming) using wavelength which was first set at 254 nm and for the first II min then changed immediately to 282 nm for the next 24 min. The recoveries of the drugs from pork and chicken muscle fortified at the level of 0.02-0.5 u g/g were 73.5~90.7%. The limit of detection of our developed method was 0.01 u g/g for each drug. |
