中文摘要 | 本研究開發以高效液相層析法同時檢驗豬飼料中Olaquindox (ODX),Carbadox( CDX), Furazolidone (FZD),Sulfathiazole (STZ), Sulfamethazine (SMT), Nalidixic Acid (NAA) ,Morantel citrate (MRT), Nitrofurazone (NFZ), Difurazone (DFZ) 等九 種合成抗菌劑。 以豬飼料 5g,用氰甲烷萃取,離心後,殘留物再以氰甲烷萃取,合併氰甲 烷液, 經三氧化二鋁 (酸性 ) 淨化, 收取濾出液經濃縮乾涸後以氰甲烷: 0.1% 磷酸 (4:6)1 ml 超音波振洫抽 30 分,經 0.45 um 濾膜過濾。 最後利用高效液相層析儀以氰甲 烷加 0.1% 磷酸為移動相, 用高純度 Silica gel 逆相 HPLC 管柱 Mightysil RP-18 GP 150 × 4.6 mm (5um) UV260 nm 及 360nm 分析, 檢体添加藥物最終濃度 0.625, 2.5 及 10 ppm 等三個濃度作分析,最終濃度為 10 ppm, UV360 nm,移動相為 80% 氰甲烷:0.1% 磷酸 (25:75)CDX, MRT,FZD 回收率平均分別為 72.5%,95.3%,100.1%,於 UV260 nm, 移動相為 80% 氰甲烷: 0.1% 磷酸 (20:80), STZ, SMT, NFZ, FZD 回收率平均分別為 66.2%,99.3 %,82.1%,95.6%,於UV360 nm,移動相為80%氰甲烷:0.1%磷酸(35:65),DFZ之 回收率平均為94.7%,而NAA及ODX回收率甚低,CDX,NFZ,MRT,FZD,SMT, STZ,DFZ線性良好,R值皆接近於1。 |
英文摘要 | A high-performance liquid chromatographic (HPLC) method for simultaneous determination of Olaquindox (ODX), Carbadox(CDX) , Furazolidone(FZD), Sulfathiazole(STZ), Sulfamethazine (SMT), Nalidixic Acid (NAA), MOrantel citrate (MRT), Nitrofurazone (NFZ), and Difurazone (DFZ) in pig feeds was developed. 5 gm of samples were extracted with acetonitrile and centrifuged. The repeated extract were combined and cleaned with Al �� O �� (acidic) column. the elute was concentrated by rotatory evaporator to dryness, and dissolved in 1 ml of acetonitrile: 0.1% phosphoric acid (4:6) for analysis by HPLC. The HPLC conditions were established on a Mightysil RP-18 GP 150 × 4.6 mm (5 um)cica-Reagent column with acetonitrile /0.1 % phosphoric acid as mobile phase and detected at UV 260 nm and UV 360 nm. The feeds samples were spiked with synthetic antimicrobial drug in 0.625, 2.5 and 10 ppm were analysed. With 80% acetonitrile:0.1% phosphoric acid (25:75) as mobile phase and detected at UV 360 nm. The average recoveries of CDX、MRT、FZD were 72.5%、 95.3%、100.1%. With 80% acetonitrile:0.1% phosphoric acid (20:80) as mobile phase and detected at UV 260 nm. The average recoveries of STZ、SMT、NFZ、 FZD were 66.2%、99.3%、82.1%、 95.6%. With 80% acetonitrile:0.1% phosphoric acid (35:65) as mobile phase and detected at UV 360 nm. The average recovery of DFZ was 94.7%. Except NAA and ODX have a poor recovery rate, CDX、 NFZ、 MRT、 FZD、 SMT、STZ、DFZ all had a good linear result. Value of R was about 1. |
