中文摘要 | 自醫院廢水中純化出6株對Stenotrophomonas maltoophilia具有溶裂作用之噬菌體。此6株噬菌體對88株臨床菌株的點試驗呈現抑菌比例分別為58%、61%、54%、51%、48%及40%。挑選其中二株øSM14b和øSM54b進行初步的genome DNA分析,其中øSM14b之DNA無法以限制酶進行切割;而噬菌體øSM54b之DNA經限制酶切割後,推估其genomic size約為50kb。將噬菌體øSM14b和øSM54b以掃描型電子顯微鏡觀察噬菌體顆粒,觀察結果顯示噬菌體øSM14b的外形為Myoviridae,而噬菌體øSM54b的外形為quasi-isometric elongated phage。在蛋白分析上,純化øSM14b約50kDa之外套蛋白進行N端定序,定序結果為SVNVTSGVQN或SSNVTSGVQN,此序列比對結果與Agrobacterium的sugar ABC transporter protein具相似性;另外亦純化øSM54b約36kDa之外套蛋白進行N端定序,定序結果為ALRLPRGIQFGFARL,並無比對到同質性較高之蛋白。 |
英文摘要 | Six lytic phages of Stenotrophomonas maltophilia were purified from hospital sewage. The inhibition efficiency of six phages to 88 clinical S. maltophilia are about 58%, 61%, 54%, 51%, 48% and 40%. Two of those phages, øSm14b and øSm54b, were selected to futher genome DNA analysis. The genomic DNA of øSm14b can not be digested by restriction enzyme, but genomic DNA of øSm54b can be digested by some restriction enzymes. The genome size of øSm54b were estimated about 50 kb. The shapes of øSm14b and øSm54b were observed by using scanning electron microscope. The result show that øSm14b and øSm54b are belong to quasi-isometric elongated phages of myoviridae. In protein sequencing, the N-terminal sequence of 50 kDa coat protein of øSm14b is determined, the result are SVNVTSGVQN or SSNVTSGVQN, and homologous compares show the sequence similar to sugar ABC transporter protein of Agrobacterium. The protein sequence ALRLPRGIQFGFARL of 30 kDa coat protein of øSm54b is also determined, but without any similarity to known protein in database. |
