中文摘要 | 利用in orgnaello DNA synthesis system 探討 nascent mtDNA的複製,我們發現新複製出的mtDNA,其□ digesting patterns與CsCl純化的mtDNA相同。而且一到四天的綠豆白化苗,除了□incorporation ability不同之外,其restriction patterns 並沒有什麼差異。依據40-55%蔗糖梯度的□ incorporation分析結果,粒線體至少可公被分成7個族群,然而由280mm吸光的結果可以發現另一個不具□ incroporation能力的MMC(muti-mitochondrial compartment)族群。我們推測粒線體的生合成含有兩個循環:一個循環不具備DNA synthesis ability (MMC),另一個循環則具備DNA synthesis ability,而且含有不同成熟度的粒線體族群(其他具有不同DNA synthesis abiltiy的粒線體族群)。 |
英文摘要 | By utilizing in organelle DNA synthesis system for analyzing the replication of nascent mtDNT, we got very clear □ digesting patterns which are identical to the □ digesting patterns of CsCl purified mtDNA and the restriction patterns were no difference among one to four days etiolated mung bean seedlings, however, the □ incorporation ability was different. According to the □ incorporation curve of 40-55% sucrose gradient after in organello DNA synthesis, mitochondria were further separated into more than seven density populations. However, one MMC (muti-mitochondria compartment) population without the □ incorporation ability was found inside the mitochondria while determined the absorbance of each fractions at 280nm. We propose that mitochondria biogenesis contains two different cycles. One of mitochondria biogenesis cycle without DNA synthesis ability (MMC), another mitochondria biogenesis cycle with DNA synthesis ability contains mature and immature mitochondria (the other mitochondria populations with different DNA synthesis abilities) during the progress of mitochondria development. |
