中文摘要 | 由於發炎反應與許多慢性疾病的病程發展有關,如何藉由飲食抑制發炎反應以減緩疾病機轉的進行,漸為許多研究者深感與趣的課題。然而,目前對抗發炎之評估常以巨噬細胞株進行體外培養,檢測添加待測樣品對巨噬細胞株分泌促發炎介質(proinflammatory mediators),例如IL(interleukin)-1β、1L-6、TNF(tumor necrosis factor)-α與nitric oxide(NO)等的影響,然而如此模式是否為理想的評估指標,值得探討。傳統中藥典記載一些中藥材具有抗發炎之功效,故本研究欲以市售之中藥復方製劑為例,探討對活化之巨噬細胞分泌發炎介質的影響,是否與初代巨噬細胞有一致性。實驗步驟分別以小柴胡湯(HCWT)、六味地黃丸(LWTHW)、雙蔘(Twinseng)與冬蟲夏草(Csm)等四種中藥製劑樣品,與同樣來自BALB/c雌鼠的RAW264.7巨噬細胞株與初代腹腔巨噬細胞,在LPS(Lipopolysaccharide)與IFN(interferon)-γ活化下,共同培養後48小時後,收集上清培養液,分析IL-1β、IL-6、TNF-α與NO等發炎介質。結果顯示,小柴胡湯、六味地黃丸與雙蔘的處理顯著抑制活化之RAW264.7細胞分泌IL-1β和IL-6的能力,只有雙蔘在低劑量有抑制NO的作用,但四種中藥樣品皆不影響細胞分泌TNF-α之能力。在初代腹腔巨噬細胞則只有六味地黃丸顯著降低細胞分泌IL-1β、IL-6和NO的作用,其它中藥樣品對於腹腔巨噬細胞分泌發炎介質物則無明顯的影響。因此,以RAM264.7巨噬細胞株探討飲食因子對促發炎介質的影響,並末與初代腹腔巨噬細胞有必然的一致性,其中以IL-1β在兩種活化的細胞培養模式中一致性較佳。四種中藥製劑中以六味地黃丸抑制IL-1β與IL-6的效應呈現一致性,顯示六味地黃丸可能較具抗發炎能力。 |
英文摘要 | Since inflammation has been reported to be associated with chronic diseases, inhibitory dietary factors may be beneficial in alleviating disease. The ability of a macrophage cell line to secrete proinflammatory mediators is often used to evaluate the anti-inflammatory effects of dietary factors. The potent anti-inflammatory properties of some Chinese herbal medicine was noted in traditional Chinese medical textbooks. Therefore, to investigate whether the effects on a macrophage cell line are consistent with those on primary macrophags derived from BALB/c mice, four Chinese herbal medicines, Hsiao-Chai-Hu-Tang (HCHT), Lui-Wei-Ti-Huang-Wan (LWTHW), Twinseng, and Cordyceps sinensis mycelia (Csm), were added to LPS/IFN-r-activated RAW264.7 or primary peritoneal macrophages. Secretions of lL-1ß, IL-6, NO, and TNF-a were measured after a 48-h incubation. Results showed HCHT, LWTHW, and Twinseng significantly inhibited IL-1ß and IL-6 secretions from activated RAW264.7 cells. Only the low dose of Twinseng had an inhibitory effect on NO secretion. TNF-a secretion was not affected by any of these four Chinese herbal medicines. In primary cell culture of peritoneal macrophages, only LWTHW significantly inhibited lL-1ß, IL-6, and NO secretion. However, the other three showed no significant effect, suggesting that the effects of dietary factors on proinflammatory mediator secretion from RAW264.7 is not necessary consistent with that on primary macrophages. Nevertheless, LWTHW, which showed significant inhibition of IL-1ß and IL-6 secretion in both the LPS/IFN-r-activated cell line and primary cells, may have an anti-inflammatory effect. |
