中文摘要 | 本試驗之主要目的為以酪胺酸、可溶性蛋白質含量及含氮物水解率之 影響評估蛋白�A對不同飼料原料蛋白質之作用。利用對酪蛋白具有相同活性 ( 1.25 units/ml ) 之兩種蛋白�A,即由 Streptomyces caespitosus 所萃取之蛋白�A 和鳳梨酵素,測定對大豆粕、魚粉、濃縮大豆蛋白及玉米麩質之水解作用。結果 顯示,不論以酪胺酸釋出量、蛋白�A作用飼料蛋白質後殘留物之氮含量或可溶性 蛋白質含量之水解速率等作比較,均顯示鳳梨酵素之效果較佳。唯由酪胺酸釋出 量之結果與後兩者之測定值評估蛋白�A對不同飼料蛋白質之作用程度,呈不一致 之趨勢,顯示,以酪胺酸釋出量多寡,並不能完全代表酵素作用後之產物型態, 因此以酪蛋白為受質及酪胺酸釋出量所測得蛋白�A活性之方法,不宜直接應用至 評估蛋白�A對飼料蛋白質作用之能力。 |
英文摘要 | The aim of this study was to evaluate the protease digestion on various feed proteins. The concentration of released tyrosine and soluble protein as well as hydrolytic rate of nitrogen compounds were used as criteria measured. Feed proteins from casein, soybean meal, fish meal, soya protein concentrate and corn gluten meal were treated with the same unit ( 1.25 units/ml ) of protease from Stroptomyces caespitosus and Bromelain. From the concentration of tyrosine released, nitrogen residue and soluble protein after different incubation times, hydrolytic activity of Bromelain was significantly higher than that Streptomyces protease. The hydrolytic activity of enzymes on different feed proteins showed the same trend in soluble protein and nitrogen residue except for concentration of tyrosine released. This showed that the amount of tyrosine released could not represent the protein residue pattern after enzyme digestion. Therefore, using casein as the substrate, the tyrosine releasing method could not be directly applied to evaluate the activity of protease on other feed proteins. |
