中文摘要 | 本試驗之目的為利用 SDS-PAGE 之圖譜探討蛋白�A對不同飼料原料 蛋白質作用之蛋白�t分佈,結果以鳳梨酵素之水解效果較 Streptomyces 蛋白�A 為佳,兩者對飼料蛋白質之蛋白�t降解效果以魚粉最佳,其次為大豆粕、濃縮大 豆蛋白,玉米麩質最差。魚粉、大豆粕及濃縮大豆蛋白經鳳梨酵素作用 30 分後, 大部份之蛋白質均被降解為小於 10 kDa 之小分子。但玉米麩質因含不易被水解 之醇溶穀蛋白,因此須 180 分後始被水解。利用 SDS-PAGE 分離飼料蛋白質, 由蛋白�t之分子量改變情形,瞭解其蛋白質組成及酵素水解效果,並得知蛋白�A 對不同組成之蛋白質作用特異性,因此得知,等活性力價之外源蛋白�A對不同飼 料蛋白質具有不同之水解效果。 |
英文摘要 | The aim of this study was to evaluate the protease digestion on various feed proteins by determining the distrbution of polypeptide pattern in hydrolyzed feed proteins. From the polyeptide pattern determined by SDS-PAGE after different incubation times, it showed that the activity of Bromelain was significantly higher than that of Streptomyces caespitosus protease. Both proteases had better hydrolyzing activity on fish meal, then followed by the soybean meal, soya protein concentrate and corn gluten meal. As fish meal, soybean meal and soya protein concentrate were treated with Bromelain for 30 minutes, the molecular weight of the polypeptides was below 10 kDa, while corn gluten meal required 180 minutes for complete digestion because of the existence of prolamins. Based on the polypeptide distribution pattern for feed proteins by SDS-PAGE, protein composition and the degree of hydrolysis were obtained. Meanwhile, there was a specificity of protease on feed protein. Therefore, feed proteins digested with the same unit of proteases, showed different hydrolytic residues. |
