中文摘要 | 利用結合牛血清白蛋白的睪固酮免疫原T-3-COM:BSA,產生的兔抗血清可成功建立睪固酮酵素免疫分析法;三隻兔血清對睪固峒的靈敏度分別為2.2±0.7、7.4±2.9與1.6±1.1pg╱assay(50μ1)。利用最靈敏的抗體做交叉反應試驗時,除與男烯二酮(4-androstene-3,17-dione)及去氫異構男性震酮(dehydroisoandrosterone)有2.4%與0.24%的交叉反應率外,與多種內源性類固醇的反應率均小於0.06%;又與多種合成的雄性素衍生物作用時發現,除1-dehydrotestosterone有73%交叉反應率外,其餘反應率均小於0.6%。實驗中的分析內變異值小於5.3%,而分析間變異值小於13.6%。本實驗亦利用雌二醇、生理食鹽水及人類絨毛膜激性腺素三種處理,觀察小鼠性腺器官生長情形與睪固酮濃度的變化。由諸精囊與睪丸、副睪重量比較,可知hCG可抭激其生長,而雌二醇具有抑制的效果。利用酵素免疫分析法分析血清與睪丸均質液中睪固酮量時,其睪固峒濃度分別為1.0±0.3、1.5±.7、13.1±1.7ng╱ml與1.1±0.9、22.4±3.9、28.2±2.6ng╱testis。體外培養孕馬血清處理後的小鼠卵巢均質液時,以加入NADPH與androstenedione的培養組有最多的睪固酮產生量。綜合上述超結果顯示,此酵素免疫分析法可成功的分析睪固酮濃度。 |
英文摘要 | An enzyme-linked immunosorbent assay (ELISA) for testosterone was established by using testosterone-3-COM: BSA as antigen. The detectability of the testosterone ELISA from three different rabbits' antisera were 2.2±0.7, 7.4±2.3, and 1.6±1.1 pg╱assay, respectively. The crossreactivity with other endogenous steroids were less than 0.06%, except for androstenedione was 2.4% and dehydroisoandrosterone whieh was 0.24%. The intraassay coefficient of variation (c.v.) was 5.3% and the interassay c.v. was 13.6% throughout the experiment. Accessory sex glands' weight and testosterone concentrations were also determined in mice under the influences of estradiol and human chorionic gonadotropin (hCG). Seminal vesicle, epididymis, and testis growth were stimulated by hCG and were inhibited by estradiol. The serum testosterone concentrations were 1.0±0.3, 1.5±0.7, and 13.1±1.7 ng╱ml(P<0.001) in estradiol treated, saline treated, and hCG treated group, respectively. The testis testosterone contents were 1.1±0.3, 22.4±3.9, and 28.2±2.6ng╱testis in estradiol treated, saline treated, and hCG treated group, respectively. Testosterone could be produced in highest concentration in pregnant mare serum gonadotropin primed mouse ovarian homogenate whe NADPH and androstenedione were added. The ELISA established in this study is useful for testosterone quantitation. |
