中文摘要 | 於5-20% C57BL/6小鼠脾臟前端背側常有黑色斑塊出現組織學觀察時,可知黑色斑塊區之紅髓中有大量含深棕色顆粒的細胞,然此深棕色顆粒之成份至今仍有爭議。為辨明此深棕色顆粒為何種物質,本試驗使用普魯士藍(Perl's Purssian blue)染色,發現此深棕色顆粒為陰性反應,排除深棕色顆粒為血鐵素(haemosiderin),以Fonata-Masson染色則為陽性反應,知此存在細胞質中的顆粒為黑色素(melanin)。再以抗CD11b抗體(rat anti-mouse CD11b antibody)與抗酥氨基酸酵素抗體(rat anti-mousetyrosinase antibody)分別作免疫染色辨識,結果發現這些黑色素顆粒存在於巨嗜細胞(macrophage),而非堆積於具生成黑色素能力的黑細胞(melanocyte)。此外,由文獻得知內臟中黑色素沈積常被認為可隨著年齡增加而消失,為檢驗C57BL/6脾臟的黑色素沉積現象是否也有此趨勢,本試驗比較5-8週齡與30-32週齡的C57BL/6小鼠產生脾臟黑色素沉積的機率,發現雖似有隨年齡有減少的趨勢,但並無統計上的差異(Fisher's exact test P=0.917)。為何脾臟黑色斑塊的出現多在前端背側,則仍待進一步的探討。 |
英文摘要 | Fifteen to twenty percent of C57BL/6 mice had natural occurring dark patches on their spleens, especially the cranial dorsal portion. However, there was argument in the constituent of the pigment, in terms of an accumulation of melanin or hemosiderin in the splenocytes. To identify whether the pigment granules in the spleen cells are made of melanin or hemosiderin, Perl's Prussian blue and Fonata-Masson stains were applied in the present histological study. The result indicates that pigment been accumulated in the spleen is constituted only by melanin (black in Perl's Prussian blue stain). The immunohistochemistry stain methods for tyrosinase and CD11b were also used to determine if the melanin containing cells in the spleen were melanocytes and/or macrophages. Since the pigment containing cells only showed positive sign to CD11b antibody, it indicated that they were macrophages. It is a nature of C57BL/6 mice that leukocytes may phagocytose significant amount of melanin in other tissues of the body, and then carry the pigment to the spleen. In addition, it has been documented in cow that organ melanosis might disappear though aging. In the present study, the occurrence of spleen melanosis in C57BL/6 mice was examined in 2 different aged-groups, 5-8 and 30-32 weeks old. However, there was no significant difference in rate of spleen melanosis between young (7/38) and aged (3/31) C57BL/6 mice. |
