中文摘要 | 本試驗使用本省過去落花生育種上常用之親本及重要育成品種(系)共30種,探討利用逢機增殖多型性DNA(RAPD)標誌進行親緣關係之判別,並與農藝性狀之結果相互比較,期由歐氏距離做為將來育種材料選定之參考,以提昇育種效率,並希望能發展出落花生種原判別鑑定之方法。由RAPD試驗結果得知,以Operon 200個引子經篩選後得11個具有多型性的引子(佔5.5%),共有212條多型性片段,其中OPE13、OPF16及OPJ17三個引子所產生之多型性片段具有與品種特性有關之分子標誌,分別位於699 bp、337 bp、及640 bp處;此三個分子標誌可作為Spanish、Valencia及Virginia三種植物型態之分辨用。另將具多型性之11個引子,其RAPD電泳圖譜進行未加權算術平均法(Un-weighted Pair-Grouping Mean Arithmetical; UPGMA)之綜合集群分析以求得樹狀圖;再以集群分析之兩群間最小距離(MD)、複相關係數(RSQ)和標準偏差(RMSSTD)等三項統計值來做為判斷集群分群個數的依據。當歐氏距離大於0.83時可分為七個族群,第一群為Spanish型之老公仔豆、臺南七號及臺南選九號;第二群為Valencia型之P1314817;第三群為臺南十一號育種系統、臺農四號育種系統及臺南十二號共9品系;第四群均為本地在來種但未運用在育種系統所用之Virginia型4品系;第五群為本地種中Spanish型之臺南白油豆二號;第六群均為本地種及引進種中之Spanish型共6品系;第七群為臺農五號育種系統及Florispan runner共6品系。由此結果可以清楚得知臺南十一號育種系統、臺農四號育種系統及臺農五號育種系統之譜系關係,與由RAPD所得資料經集群分析結果之分群結果頗為符合,並與植物型分類結果相符合;依上述結果推論,落花生種原若使用本試驗所得之引子進行歐式距離分析以及親緣關係之探討,應屬相當可行。 |
英文摘要 | This experiment was conducted by using randomly amplified polymorphic DNA marker (RAPD marker) analysis to evaluate the genetic relationship of peanut germplasm. There are totally 30 varieties (lines), which were frequently supplied as the source of peanuts and main bred varieties (lines) in breeding program in Taiwan were analyzed. The results of RAPD were compared with the agricultural traits. The correlation coefficient get from RAPD can be used as the index to select the varieties (lines) for breeding program to increase the genetic gains. This method also can be developed as a protocol for identification of peanut germplasm. There were totally 200 operon decamer primers was used to screen the polymorphism response, only 11 primers (5.5%) show positive and produced 212 reproducible and effective bands.Three identical bands produced by OPE13, OPF16 and OPJ17 at 699 bp, 3347 bp and 640 bp respectively can be look as marker bands for botanical type identification. In this way three botanical type of Spanish type, Valencia type and Virginia type can be distinguish by three marker bands. The RAPD profiles of 11 polymorphic primers were converted into the matrix for Euclidean distances based on Dice's coefficient to proceed with the cluster analysis by UPGMA (Unweighted Pairj-Grouping Mean Arithmetical). The numbers of cluster are based on the MD, RSQ and RMSSTD obtained from cluster analysis. The 30 varieties (lines) could be divided into 7 main clusters when the Euclidean distances were greater than 0.83. The first cluster contained 3 varieties of Spanish type named Lau-Kung-Tzu-Tou, Tainan No. 7 and Tainann selected No.9. The second cluster only has one variety PI314817 (Valencia type).The third cluster contained 9 varieties, which also belong to the breeding systems of Tainan No.11, Tainung No.4 and Tainan No.12. The fourth cluster contained 4 native varieties of Virginia type which were not used in the breeding programs. The fifth cluster signally contained Tainan white oil bean No.2, which is a native Spanish type variety. The sixth cluster contained 6 varieties which were used in Tainung No.5's program and Florispan runner. According to the result of cluster analysis of TAPD significantly revealed the pedigree diagrams of the breeding programs of Tainan No.11, Tainung No.4 and Tainung No.5, and also closely related to plant botanical types. The results could be deducted that 11 selected primers in this study can be effectively and reliably used for studies of genetic distances and the genetic relationship of peanut germplasm. |
